The full-length genome sequence of a novel mitovirus from Botryosphaeria dothidea, the causal agent of pear ring rot disease

Abstract: Here, we describe a novel mycovirus, Botryosphaeria dothidea mitovirus 2 (tentatively designated as BdMV2), isolated from Botryosphaeria dothidea FJ strain causing pear ring rot disease in Fujian Province, China. The complete genome nucleotide sequence of BdMV2 is 2538 nt in length and contains a single 2070 nt open reading frame (ORF) encoding a putative RNA-dependent RNA polymerase (RdRp) of 689 amino acid (aa) using fungal mitochondrial genetic code. BLASTp analysis revealed that the RdRp of BdMV2 shares 28… Show more

“…This feature was found in the study by Botella and collaborators (2022) [ 106 ], who described narnaviruses in the fungus Phytophthora palustris , viruses that are related to the traditional positive-coding monopartite, included in the genus Narnavirus . Also of note, Kinsella and collaborators (2022) [ 107 ] identified narnaviruses in the hosts R. oryzae and A. lentulus , which are pathogenic fungi, and Zou et al (2021) [ 108 ] analyzed the fungus Botryosphaeria dothidea , which is the causal agent of the disease pear ring rot. The viral contig NVc is the first narnavirus detected in Ceratocystis cacaofunesta.…”

“…Then, adapter-related reads, reads for which basic information could not be determined, low-quality reads, and host-related reads were further removed from the raw reads. Subsequently, the cleaned reads were spliced and compared to GenBank, NR, and CDD databases using Trinity and DIAMOND software, respectively [33][34][35]. Finally, we obtained viral RNA sequences.…”

“…So far, 18 mycoviruses have been described from various strains of B. dothidea: Ten of them are dsRNA viruses from four families ( Chrysoviridae , Partitiviridae , Totiviridae , and the proposed Botybirnaviridae), and two are unassigned [ 25 , 26 , 27 , 28 , 29 , 30 , 31 , 32 , 33 ]. The remaining eight viruses are +ssRNA viruses in the families Alphaflexiviridae [ 34 ], Fusariviridae [ 35 , 36 ], Mitoviridae [ 37 , 38 , 39 ] and Botourmiaviridae [ 40 , 41 , 42 ]. Some of these mycoviruses have been identified from hypovirulent strains of B. dothidea , suggesting that they might have a negative impact on the fungal host virulence [ 25 , 28 , 32 , 34 ].…”
