The Fuchsin-Aldehyde Reaction on the Endo Medium

Abstract: Endo (1904) suggested the use of a medium of the following composition for the differentiation of B. typ wu from B. colilike organisms:

“…The decolorization, which takes place if the plate is allowed to stand long enough, is due to such alkalis as amnmonia and amines, which are formed from the peptone and the beef extract. DeBord (1917) performed some experiments to test the effect of acid alone, combinations of acid and formaldehyde, and formaldehyde alone on decolorized basic fuchsins as well as on Endo agar. He thinks the reaction is an acid-aldehyde reaction and that the presence of an aldehyde is necessary to it.…”
