The front line of research into immunoglobin G4-related disease - Do autoantibodies cause immunoglobin G4-related disease?

Abstract: IgG4-related disease (IgG4-RD) is a novel clinical disease entity characterized by elevated serum IgG4 concentration and tumefaction or tissue infiltration by IgG4-positive plasma cells. Although IgG4-RD is attracting strong attention as a new clinical entity, the pathogenesis of IgG4-RD and the roles of IgG4 are still unknown. Recently, self-antigens including Laminin 511 E8, Galectin-3 and Annexin A11, have been reported from one to the next as autoantigens which may be involved in the pathogenesis of IgG4-R… Show more

“…These findings can also be supported by a recently reported co‐occurrence of IgG4‐RH and a craniopharyngioma 42 . Genetic predisposition and presence of autoantibodies have already been suggested as mechanisms responsible for the development of IgG4‐RD 43‐45 …”

“…The PD-1/PD-L1 pathway also contributes to the interaction between T follicular helper cells and plasmablasts [9]. One possible mechanism is that the inhibition of PD-1/PD-L1 pathway enhances T follicular helper cells to promote the growth, differentiation, and class switching of B cells to IgG4 [11,12]. Another possibility is that there is an association between pulmonary adenocarcinoma and IgG4-related disease.…”

“…Numerous self-antigens have been reported as potential autoantigens responsible for the pathogenesis of IgG4-RD [31]. However, it is unclear whether IgG4 antibodies are pathogenic in IgG4-RD or merely represent a regulatory response to another primary autoimmune or allergic process [1,19,[31][32][33][34][35]. In an experimental animal model, J Investig Allergol Clin Immunol 2021; Vol.…”
