The Fragile X Syndrome Single Strand d(CGG)n Nucleotide Repeats Readily Fold Back to Form Unimolecular Hairpin Structures

Abstract: Expansion of a d(CGG)nFragile X syndrome is an inherited, X-linked dominant mental retardation disorder affecting about one male in 1500 and one female in 2500 (Webb, 1989;Richards and Sutherland, 1992). This syndrome is frequently associated with a folatesensitive fragile site, Xq27.3, on chromosome X of cells of affected males (Sutherland, 1977). The fragile X syndrome is characterized by a substantial expansion of a d(CGG) trinucleotide repeat located in the 5Ј-untranslated region of a housekeeping gene, FM… Show more

“…Formation of Tetraplex DNA Structures-The trinucleotide repeat sequence d(CGG) n has been shown to readily fold into hairpin structures (31)(32)(33)(34) and to assemble into tetrahelical formations (25)(26)(27). Tetraplex structures of d(CGG) n are bonded by guanine-guanine non-Watson-Crick hydrogen bonds and are stabilized by monovalent alkali cations (25)(26)(27).…”

“…Following UV cross-linking, this form also migrated rapidly in a denaturing 8 M urea, 12% polyacrylamide gel (9). The folded DNA structure was resistant to methylation with dimethyl sulfate, indicating its stabilization by Hoogsteen hydrogen bonds (49). To prepare GЈ2 TeR DNA, a bimolecular tetraplex form of oligomer TeR, 10 M TeR2 DNA in TE buffer was heated at 95°C for 2 min followed by incubation of the DNA at 37°C for 20 h in the presence of 1.0 M KCl.…”

“…Aside from being potential ''anti-cancer knots,'' G-quartets are also linked to site-specific genetic recombination in immunoglobulin switch regions (9), the dimerization of HIV RNA (10), and L1 retroposition (11). More recently, DNA quadruplexes have been associated with the promoter regions of DNA, such as the triplet repeat sequence that causes fragile-X syndrome (12)(13)(14), the retinoblastoma susceptibility genes (15), and the chicken ␤-globulin gene (16), raising tantalizing possibilities of controlling gene expression with G-quartets.…”
