Bronchial asthma, one of the most common allergic diseases, is characterized by airway hyperresponsiveness (AHR), inflammation, and remodeling. The anti-oxidant flavone aglycone diosmetin ameliorates the inflammation in pancreatitis, but little is known about its impact on asthma. In this study, the effects of diosmetin on chronic asthma were investigated with an emphasis on the modulation of airway remodeling in BALB/c mice challenged with ovalbumin (OVA). It was found that diosmetin significantly relieved inflammatory cell infiltration, goblet cell hyperplasia, and collagen deposition in the lungs of asthmatic mice and notably reduced AHR in these animals. The OVAinduced increases in total cell and eosinophil counts in bronchoalveolar lavage fluid were reversed, and the level of OVA-specific immunoglobulin E in serum was attenuated by diosmetin administration, implying an anti-Th2 activity of diosmetin. Furthermore, diosmetin remarkably suppressed the expression of smooth muscle actin alpha chain, indicating a potent anti-proliferative effect of diosmetin on airway smooth muscle cells (ASMCs). Matrix metallopeptidase-9, transforming growth factor-β1, and vascular endothelial growth factor levels were also alleviated by diosmetin, suggesting that the remission of airway remodeling might be attributed to the decline of these proteins. Taken together, our findings provided a novel profile of diosmetin with anti-remodeling therapeutic benefits, highlighting a new potential of diosmetin in remitting the ASMC proliferation in chronic asthma.

The present study aims to improve the biological activities of luteolin by complexation with manganese(II).UV-visible spectroscopy, infrared spectroscopy, thermogravimetric analysis and elemental analysis were adopted to assess the relevant interaction of luteolin and manganese(II) ions and the chelation sites. The antioxidant activity, hypoglycemic effect and antimicrobial activity of luteolin-manganese(II) complex with respect to its parent luteolin and the inhibition effect of which on xanthine oxidase were investigated and compared. The spectroscopic data indicated that luteolin reacts with manganese(II) cations through the chelation sites of 5-hydroxy and 4-carbonyl in two luteolin molecules. Antioxidant and antibacterial activity were enhanced after the complexation of manganese(II) cations with luteolin. An inhibition effect assay found that luteolin and luteolin-manganese(II) complex reversibly inhibited xanthine oxidase in a competitive manner. Luteolin-manganese(II) complex had a more remarkable hypoglycemic effect than luteolin by increasing the glucose consumption in liver tissue.

Purpose: To improve the hydrophobicity of diosmetin, the complex of diosmetin and lecithin was prepared and its physicochemical properties were characterized in this study. Methods: The complex of diosmetin and lecithin was prepared by solvent method. The physicochemical properties of the complex were investigated by ultraviolet-visible spectrometry (UV/Vis), infrared spectrometry (IR), scanning electron microscopy (SEM), differential scanning calorimetry (DSC) and x-ray diffractometry (XRD). Results: Based on UV and IR analysis, there was no significant difference between the physical mixture and the complex while SEM showed that diosmetin in the complex did not present a crystal appearance but rather was dispersed in lecithin. DSC thermograms of the complex mainly showed that the presence of lecithin caused the characteristic endothermal peaks of diosmetin to disappear while x-diffractogramsshowed that the crystalline peaks of diosmetin was absent in the complex. Conclusion:The complex is held together by non-covalent-bond, and thus a new compound was not formed. Diosmetin in the complex is molecularly dispersed in the lecithin matrix.
