The precise balance of Th1, Th2, and Th17 cytokines is a key factor in successful pregnancy and normal embryonic development. However, to date, not all humoral factors that regulate and influence physiological pregnancy have been completely studied. Our data here pointed out cyclophilin A (CypA) as the adverse pro-inflammatory factor negatively affecting fetal development and associated with pregnancy complications. In different mouse models in vivo, we demonstrated dramatic embryotoxicity and teratogenicity of increased CypA levels during pregnancy. Using generated transgenic models, we showed that CypA overexpression in fetal tissues induced the death of all transgenic fetuses and complete miscarriage. Administration of recombinant human CypA in a high dose to pregnant females during fetal organogenesis (6.5–11.5 dpc) exhibited teratogenic effects, causing severe defects in the brain and bone development that could lead to malformations and postnatal behavioral and cognitive disorders in the offspring. Embryotoxic and teratogenic effects could be mediated by CypA-induced up-regulation of M1 macrophage polarization via activation of the STAT1/3 signaling pathways. Here, we propose secreted CypA as a novel marker of complicated pregnancy and a therapeutic target for the correction of pregnancy complications.

This study aimed to determine the expression of microRNA (miRNA or miR)-378 in peripheral blood from patients with pregnancy-induced hypertension (PIH), and to try to understand its effect on vascular endothelial injuries induced by PIH. The study included 33 PIH patients. Quantitative real-time polymerase chain reaction was used to determine the expression of miR-378 in peripheral blood and NK cells. NK cells were transfected with miR-378 mimics and its negative control. Bioinformatics was used to predict the potential target gene of miR-378. Flow cytometry and Western blotting were performed to identify changes in the expression of CD226 protein. By co-culturing HUVECs with NK cells, the effects of miR-378 and CD226 on the proliferation, migration and apoptosis of HUVECs were evaluated. The results showed that the expression of miR-378 in peripheral blood was down-regulated with the progression of PIH. The expression of miR-378 was reduced in innate immune NK cells from peripheral blood of PIH patients. miR-378 negatively regulated the expression of CD226 in NK cells, and CD226 is a target gene of miR-378. miR-378 inhibited the expression and secretion of soluble CD226 by NK cells. miR-378 regulated the proliferation, migration and apoptosis of HUVECs by affecting the levels of soluble CD226 expressed by NK cells. The present study demonstrates that miR-378 is down-regulated in peripheral blood and NK cells from patients with PIH, and negatively correlated with the development of PIH. miR-378 protects HUVECs from injuries by soluble CD226 through down-regulating the expression of soluble CD226 in NK cells.

Introduction: Cyclophilin A was reported to be increased in the serum of mothers with preeclampsia, and is implicated in its pathogenesis. This study aimed to determine the expression of cyclophilin A in the placenta of mothers with and without hypertension, and to correlate its expression with maternal complications and adverse perinatal outcomes. Materials and Methods: This study consisted of a total of 70 cases (35 cases of mothers with hypertension, and 35 normotensive mothers as a control). Cyclophilin A immunohistochemistry was performed on a paraffin-embedded tissue section of placenta submitted at full thickness in order to evaluate the expression in fetal endothelial cells, cytotrophoblasts, syncytiotrophoblasts, maternal endothelial cells and decidual cells. The cyclophilin A expression was scored as weak, moderate or strong intensity. Results: The hypertensive group was more likely to have preterm deliveries (p < 0.0001), caesarean sections (p < 0.0001), and infants admitted to the intensive care unit (p < 0.001). Fifty-one percent of the fetal endothelial cells and cytotrophoblasts expressed cyclophilin A in the hypertensive group, compared to only 28.6% in the normotensive group. However, the difference was not statistically significant (p = 0.086). Conclusion: We found no significant difference in placental cyclophilin A expression between hypertensive and normotensive mothers. There was also no difference in expression in mothers with and without maternal complications and adverse perinatal outcomes.
