One health is an emerging conceptual approach geared to harmonize the activities of the public health, veterinary services, and extension services within a single operative structure. Brucellosis is an important zoonosis worldwide, mostly involving nomadic populations but may often affect transboundary animal management and exotic domesticated animal farming such as camels and buffalo. Here, we provide contemporary knowledge on the disease and its causative agent, a Gram-negative bacteria belonging to the genus Brucella. Further, because of the zoonotic importance, we emphasize the need to assign a national reference laboratory for the disease and discuss how this would integrate into a “One Health” system. Brucella vaccines are live attenuated strains possessing the smooth phenotype, and vaccination, therefore, hampers the ability to maintain a national surveillance program due to concerns regarding the false positive vaccine-induced responses. In order to overcome these failings, we developed a combined approach based on rapid screening of mass numbers of serum samples by the fluorescence polarization assay, a cost-effective and accurate method, and confirmation of the true positive reactors by the complement fixation test, a highly specific method that is less sensitive to vaccine-induced antibodies. We demonstrate how, despite the high vaccination coverage of the small ruminant population in Israel, our results proved to be effective in discriminating between vaccinated and infected animals. The speed and accuracy of the method further justified immediate declaration of 37% of flocks as cleansed from brucellosis, thus reducing the burden of repeated tests among this population.

France has been officially free of bovine brucellosis since 2005. Nevertheless, in 2012, as the source of two human cases, a bovine outbreak due to B. melitensis biovar 3 was confirmed in the French Alpine Bargy massif, due to a spillover from wild, protected Alpine ibex (Capra ibex). In order to reduce high Brucella prevalence in the local ibex population, successive management strategies have been implemented. Lateral flow immunochromatography assay (LFIA) was thus identified as a promising on-site screening test, allowing for a rapid diagnosis far from the laboratory. This study compared a commercial LFIA for brucellosis diagnosis with the WOAH-recommended tests for small ruminants (i.e., Rose Bengal test (RBT), Complement fixation test, (CFT) and Indirect ELISA, (iELISA)). LFIA showed the same analytical sensitivity as iELISA on successive dilutions of the International Standard anti-Brucella melitensis Serum (ISaBmS) and the EU Goat Brucella Standard Serum (EUGBSS). Selectivity was estimated at 100% when vaccinated ibex sera were analyzed. When used on samples from naturally infected ibex, LFIA showed high concordance, as well as relative sensitivity and specificity (>97.25%) in comparison with RBT and CFT. This work shows high reliability and ensures a better standardization of LFIA testing for wild ruminants.

Background Brucellosis is a serious zoonosis disease that frequently causes significant economic loss in animal husbandry and threatens human health. Therefore, we established a rapid, accurate, simple and sensitive fluorescent immunochromatographic strip test (ICST) based on quantum dots (QDs) for detection the antibodies of Brucella infection animals serum. Results The test strips were successfully prepared by quantum dot fluorescent microspheres (QDFM) as tracers, which were covalently coupled to an outer membrane protein of Brucella OMP22. The outer membrane protein OMP28 and monoclonal antibodies of OMP22 were separately dispensed onto a nitrocellulose membrane as test and quality control lines, respectively. The critical threshold for determining negative or positive through the ratio of the fluorescent signal of the test line and the control line (HT / HC) is 0.0492. The repeatability was excellent with an overall average CV of 8.78%. Under optimum conditions, the limit of detection was 1.05 ng/mL (1:512 dilution). With regard to the detection of brucellosis in 150 clinical samples, the total coincidence rate of ICST and Rose Bengal plate test (RBPT) was 97.3%, the coincidence rate of positive samples was 98.8%, the coincidence rate of negative samples was 95.3%, the sensitivity of RBPT is 1:32, and no cross reaction with the sera of other related diseases was observed. Conclusion In our present study, the QDFM has promising application for on-site screening of brucellosis owing to its high detection speed, high sensitivity, high specificity and low cost.
