The extracellular matrix glycoprotein tenascin-R regulates neurogenesis during development and in the adult dentate gyrus of mice

Abstract: Abnormal generation of inhibitory neurons that synthesize c-aminobutyric acid (GABAergic) is characteristic of neuropsychological disorders. We provide evidence that the extracellular matrix molecule tenascin-R (TNR) -which is predominantly expressed by a subpopulation of interneuronsplays a role in the generation of GABAergic and granule neurons in the murine dentate gyrus by regulating fate determination of neural stem or progenitor cells (NSCs). During development, absence of TNR in constitutively TNR-defic… Show more

“…Some genes ( SLIT1 , TNR , FAM181B , IFT172 , PPM1G , SNX17 , and PREB ) associated with neural development [38, 39, 40, 41, 58, 59, 60, 61, 62, 63] were also found in the top 20 significant windows for both traits. Costa et al [10] also detected genes related to neural development that were associated with reproductive traits.…”

“…In the olfactory bulb, the related gene Tnr attracts the neuroblasts into the periglomerular networks (Saghatelyan et al, 2004). The latter glycoprotein Tnr also seems of importance in the neurogenic zone of the adult hippocampus, since the Tnr knock-out mouse line shows an increased number of GABAergic neurons, a reduced expression of b1-integrin, an enhanced activation of p38 MAPK and an augmented expression of the GABAergic specification gene Ascl1 (Xu et al, 2014). Interestingly, Tnc is not only found in the NSC niche, but also in stem cell niches of other organs and hence emerges as a constitutive element in various stem cell microenvironments (Chiquet-Ehrismann et al, 2014).…”

“…TN-R has also a modular structure like TN-C, but forms trimers instead of hexamers (Joester and Faissner, 2001). TN-R interacts with Integrins, for example β1 Integrins (Xu et al, 2014), Contactin/F3/F11, Phosphacan (Jones and Jones, 2000a), and Myelin-associated glycoprotein (MAG; Yang et al, 1999). TN-R is expressed after spinal cord injury (Apostolova et al, 2006) and in the lesioned optic nerve by an increased number of cells (Becker et al, 2000).…”
