The Expression of Metalloproteinase-2, −9, and −14 and of Tissue Inhibitors-1 and −2 Is Developmentally Modulated During Osteogenesis In Vitro, the Mature Osteoblastic Phenotype Expressing Metalloproteinase-14

Abstract: During osteogenesis, in vitro, of tibial-derived rat osteoblasts (ROB) and derived clones, changes occur in the interactions of mature osteoblasts with the endogenous extracellular matrix (ECM) and these culminate in the formation of tridimensional nodules, which become sites of mineral deposition. We investigated if these changes might be mediated by remodeling of ECM, and we focused our study on the neutral metalloproteinases (MMPs), known agents of matrix remodeling, and on their tissue inhibitors (TIMPs). … Show more

“…Enzyme assays and Western blot techniques have revealed the presence of MMP-2 as well as MMP-9 in paw tissue extracts prepared from rats with AIA. Similarly, expression of several MMPs (including MMP-9, MMP-8, MMP-13, and MMP-14) has been observed in, or immediately adjacent to, the epiphyseal growth plates of rats (31)(32)(33)(34)(35). In addition, MMP-3 is reported to be involved in the activation of transforming growth factor ␤1 in rat growth plate chondrocytes (36).…”

“…Increase of IL-11, an important factor in osteogenesis (23), was shown after 2 hours of static tensional force as well (Table 1). MMPs and tissue inhibitors of metalloproteinases (TIMPs) are similarly important in new bone formation (24). The expression of MMPs and TIMPs in PDL cells was altered in response to static tensional force.…”

“…During osteogenesis, MMP-2 is expressed in the osteoblast while TIMP-1 is related to its maturation (24). Therefore, orchestrated expression of MMPs and TIMPs might be significant, as shown in the formation of new bone (24) and tooth eruption (26).…”

“…17 Osteoblast survival and apoptosis 4,19 depend on endogenous growth factors, and often associated with the ECM. Progression of cell differentiation and/or the maturation of the osteogenic ECM require developmentally regulated expression of endogenous metalloproteinases 11 (Manduca et al, unpublished data).…”
