The expression of genes related to adipocyte differentiation in pigs1

Abstract: The purpose of this study was to detect differential expression of genes related to adipocyte differentiation in pigs by suppression subtractive hybridization. Adipocytes and stromal vascular cells (a fraction containing preadipocytes) from pig adipose tissue were isolated for mRNA extraction. The cDNA from preadipocytes was subtracted from the cDNA from adipocytes. The subtracted gene fragments were cloned into pGEM-T Easy TA cloning vector. We selected 384 clones for gene sequence determination and for furth… Show more

“…Rosi = 1 M of rosiglitazone treatment. et al, 2004;Wang et al, 2006). The densitometric value for an individual transcript in a sample lane was normalized to the densitometric value for the glyceraldehyde-3-phosphate dehydrogenase (GAPDH) mRNA in the same lane.…”

“…The results of the present investigation indicate that ACE inhibitors increase the expression of CRBP1. However, because this protein would be expected to have greater effects on adipocytes, which in addition to being involved in retinoid storage and metabolism express high levels of ACE (Wang et al, 2006), the majority of our study investigated its effects either in adipocytes or in vivo. Studies in rats indicate that 15 to 20% of the total retinoid content in the body can be stored in adipose tissue (Tsutsumi et al, 1992), and CRBP1 expression in adipocytes is regulated by retinoic acid, dexamethasone, and tri-iodothyronine (Okuno et al, 1995) , an unrelated control siRNA (each 50 pmol) or transfection reagent alone (without siRNA) on the expression of a histidinetagged CRBP1 in human embryonic kidney 293 cells.…”

“…A transgenic mouse lacking a functional ATM gene has early embryonic lethality (Huber et al 2004). The porcine ATM gene was cloned (Wang et al 2006) and is expressed ubiquitously in pig adult tissues with the greatest expression in tissues with the most proliferation (Rogatcheva et al 2007). Perhaps, the rapid increase in blastocyst cell number required tighter control over DNA replication and the cell cycle, thus requiring increased ATM.…”
