The Exosome Total Isolation Chip

Abstract: Circulating tumor-derived extracellular vesicles (EVs) have emerged as a promising source for identifying cancer biomarkers for early cancer detection. However, the clinical utility of EVs has thus far been limited by the fact that most EV isolation methods are tedious, nonstandardized, and require bulky instrumentation such as ultracentrifugation (UC). Here, we report a size-based EV isolation tool called ExoTIC (exosome total isolation chip), which is simple, easy-to-use, modular, and facilitates high-yield … Show more

“…Compared with ultracentrifugation, this technique achieved moderately lower separation throughput and exosomes yield. To sort different‐sized EVs from the sample, a multiple filtrations microfluidic chip, called ExoTIC, was developed to separate a subpopulation of EVs (Figure B) . ExoTIC was designed as a modular unit that consisted of membranes with different pore sizes (e.g., 200, 100, 50, and 30 nm).…”

“…This method co-isolates small EVs and some large EVs with high throughput, high yield and low levels of protein contaminants [90]. Liu et al demonstrated a size-based modularised Mf-F system called ExoTIC, which is a modular platform with the ability to isolate EV subpopulations based on size [91] (Figure 2(g)). …”

“…Liu et al reported that three miRNAs, miR-23b-3p, miR-10b-5p, and miR-21-5p, appeared to be promising prognostic biomarkers in a patient with NSCLC. They isolated exosomes from blood from 196 patients and used qPCR assay to determine the enrichment of miRNAs (Liu, Vermesh, et al, 2017; Liu, Yu, et al, 2017). …”

“…Moreover, the researchers demonstrated that ExoTIC can sort a heterogeneous population of cancer cell exosomes based on size. They also tested ExoTIC to isolate exosomes from clinical samples, including plasma, urine, and lavage, demonstrating the device’s broad applicability to cancers and other diseases (Liu, Vermesh, et al, 2017; Liu, Yu, et al, 2017). …”
