The Exocyst Complex Regulates Free Fatty Acid Uptake by Adipocytes

Abstract: The exocyst is an octameric molecular complex that drives vesicle trafficking in adipocytes, a rate-limiting step in insulin-dependent glucose uptake. This study assessed the role of the exocyst complex in regulating free fatty acid (FFA) uptake by adipocytes. Upon differentiating into adipocytes, 3T3-L1 cells acquire the ability to incorporate extracellular FFAs in an insulin-dependent manner. A kinetic assay using fluoresceinated FFA (C12 dodecanoic acid) uptake allows the real-time monitoring of FFA interna… Show more

“…Several genes that are cis -regulated in the muscle participate in lipid or carbohydrate metabolism, e.g. HIV-1 Tat Interactive Protein 2 ( HTATIP2 ) [14], exocyst complex component 7 ( EXOC7 ) [15] acyl-CoA dehydrogenase short chain ( ACADS ) [16], insulin degrading enzyme ( IDE ) [17], solute carrier family 38 member 9 ( SLC38A9 ) [18], and family with sequence similarity 3 member C ( FAM3C ) [19]. With regard to the liver, hydroxysteroid dehydrogenase like 2 ( HSDL2 ) [20] and lipase A ( LIPA ) [21] genes have also important roles in lipid metabolism.…”

“…5b). While CHCHD3 maintains the structural integrity of mitochondrial cristae 28 , EXOC4 is a component of the exocyst complex involved in the secretion of insulin 29 , as well as lipid and glucose uptake in response to insulin signaling 30 . SNP 3, the most significant variant within the suggestive subset of loci, was closest to motilin ( MLN; Supplementary Fig.…”
