The Evolutionary History of TLR4 Polymorphisms in Europe

Abstract: Infections exert important evolutionary pressures shaping the human genome, especially on genes involved in host defense. A crucial step for host defense is recognition of pathogens by pattern recognition receptors on innate immune cells, among which Toll-like receptor 4 (TLR4) is one of the best known. Genetic variation in TLR4 (Asp299Gly, Thr399Ile) has been recently described. Haplotype frequencies of these polymorphisms differ among African, Asian and European populations, suggesting evolutionary pressures… Show more

“…The preponderance of research in sepsis has focused on dissecting roles of the immune cells, innate immune regulation, cytokines, and coagulation factors in response to varying infectious and inflammatory mediators [11, 18]. This meta-analysis mainly focused on the associations between CD14-159C/T polymorphism and susceptibility to sepsis or sepsis-related mortality.…”

“…Several studies tried to establish an association between CD14 polymorphism and sepsis [10, 11]. However, varied conclusions have been obtained in different studies.…”

“…Our data are in agreement with previous reports showing that mutations and susceptibility alleles for autoimmune diseases such as celiac disease and type 1 diabetes have been maintained in human populations because of past positive selection and adaptation. [30][31][32] Immune response and reproduction are believed to be the main selective pressures that have shaped human evolution. 30,33 Exploring further, the geographic distribution of variants in these two genes will provide a unique model to dissect evolutionary constraints on genes with roles in both immune responses and reproduction.…”

“…In addition, the TLR4 polymorphism Asp299Gly has been shown to be associated with otitis-prone condition in human studies 21 and polymorphisms in the TLR4 gene (Asp299Gly and Thr300Gly) are present in 10% of the Caucasian population and have been shown to be associated with numbers health risks, i.e. gram negative sepsis, premature birth 22 . This mouse therefore offers a model in which to characterize the impact of chronic middle ear inflammation on key homeostatic mechanisms in the cochlea and middle ear.…”
