The Endoplasmic Reticulum Provides the Membrane Platform for Biogenesis of the Flavivirus Replication Complex

Abstract: The cytoplasmic replication of positive-sense RNA viruses is associated with a dramatic rearrangement of host cellular membranes. These virus-induced changes result in the induction of vesicular structures that envelop the virus replication complex (RC). In this study, we have extended our previous observations on the intracellular location of West Nile virus strain Kunjin virus (WNV KUN ) to show that the virus-induced recruitment of host proteins and membrane appears to occur at a pre-Golgi step. To visualiz… Show more

“…In our experiments, 5-month-old analyzed by means of immunofl uorescence and confocal microscopy 24 h p.i. Cells showing cytoplasmic cumuli of dsRNA intermediates, which are markers of WNV replication ( 3,5 ), as well as cells positive for WNV E glycoprotein, were observed in infected cultures ( Fig. 4A ), supporting the ability of WNV to replicate in these fi broblast lines.…”

“…5A, B ). To further analyze this point and to test whether high SM levels had the same effect in other cell lines, we added SM to Vero cells, which are a widely used model for WNV infection ( 3,5 ). The addition of SM signifi cantly increased SM content of Vero cells (5.95 ± 1.23 nmol/mg and 12.44 ± 2.34 nmol/mg of protein in control and treated cells, respectively) ( Fig.…”

“…As reported for other flaviviruses, the mechanism of penetration into the cytoplasm must be initiated by the fusion of the viral E with the membranes of the cellular endosomes from the host cell, a process triggered by the acidic pH inside cellular endosomes. 38 The implication of cellular cofactors in membrane rearrangements induced by USUV has been analyzed by transmission electron microscopy after USUV infection, showing, as previously reported for other flaviviruses, 39 the presence of vesicle packets with electron-dense virions 40 ( Figure 3B). Lipid requirement analyses for USUV replication have confirmed fatty acid and other cellular lipids as essentials for replication, since treatment with acetyl coenzyme A carboxylase inhibitors impaired USUV multiplication.…”

“…The viral genome is translated as a single polyprotein that is cleaved by host and viral proteases into three structural (C, prM, and E) and seven nonstructural (NS1, NS2A, NS2B, NS3, NS4A, NS4B, and NS5) proteins (4,5). The structural proteins are components of viral particles and the nonstructural proteins form the replication complex that is essential for replication of viral RNA (6,7). The viral particles assemble and bud into the ER to form immature particles (8).…”
