The Emerging Role of Sigma Receptors in Pain Medicine

Abstract: Sigma receptors are protein chaperones with the unexpected characteristic of being activated by ligand binding. As such, they represent intriguing new targets for potential drug development. As a protein chaperone, these "receptors" escort proteins from the endoplasmic reticulum to their destinations and act to correct misfolded proteins. The two subtypes of sigma receptors, named σ1 and σ2, are markedly distinct from each other. Agonists and antagonists at these receptors show promise as new drug targets, add… Show more

“…The role of genetics, the microbiome, and other factors may be the next radical paradigm shift in pain medicine [91]. New therapeutic targets are being discovered [92]. There are many exciting new horizons in pain care, as well as revitalization efforts for the mainstays of pain care.…”

“…This property aids in the proper transport and folding of proteins, a property that is required for proper functioning. Such an activity would have broad therapeutic utility in situations of dysregulated or disrupted physiological pathways or excursions from baseline activity, such as is hypothesized to occur in neuropathic pain [2,3]. Here, we review some recent publications that provide examples of the analgesic activity of some S2Rselective compounds.…”

“…As described in recent reviews a valuable role of sigma receptors is as mediators of pain-relieving mechanisms, and therefore they provide pharmacologic targets for the discovery and development of novel analgesic agents [ 2 , 3 ]. Conventional analgesic therapy and medications suffer from well-known challenges, such as insufficient clinical efficacy, worrisome adverse effects, or problems with abuse.…”
