The EH and SH3 domain Ese proteins regulate endocytosis by linking to dynamin and Eps15

Abstract: A.S.Sengar and W.Wang contributed equally to this workClathrin-mediated endocytosis is a multistep process which requires interaction between a number of conserved proteins. We have cloned two mammalian genes which code for a number of endocytic adaptor proteins. Two of these proteins, termed Ese1 and Ese2, contain two N-terminal EH domains, a central coiled-coil domain and five C-terminal SH3 domains. Ese1 is constitutively associated with Eps15 proteins to form a complex with at least 14 protein-protein inte… Show more

“…The Intersectin adaptor protein is thought to function in clathrin-dependent endocytosis by recruiting endocytosis machinery to clathrin-coated pits (Yamabhai et al, 1998;Sengar et al, 1999;Simpson et al, 1999;Koh et al, 2004;Marie et al, 2004). The C. elegans genome contains a single Intersectin orthologue, called ITSN-1, which is most similar to Drosophila dynamin-associated protein of 160 kDa (Dap160) and the short isoform of human ITSN1 ( Figure 5A).…”

“…One unique feature of WNK proteins is that they contain multiple PXXP motifs known for binding SH3 domains. A recent study (He et al, 2007) reports that WNK1 and 4 stimulate CCVmediated endocytosis of ROMK by recruiting intersectin, a multimodular endocytic scafford protein containing five SH3 domains (Sengar et al, 1999). The effects of WNK1 and 4 on NCC also occur through changes of membrane expression (Yang et al, 2003;Cai et al, 2006).…”

“…Intersectin is a scaffolding protein that has five SH3 domains (SH3A-SH3E), which are sites for protein-protein interactions (Roos and Kelly, 1998;Yamabhai et al, 1998;Sengar et al, 1999). Through its protein interactions, intersectin has been demonstrated to function in endocytosis (Yamabhai et al, 1998;Simpson et al, 1999) and the regulation of actin dynamics , and it has also been implicated in exocytosis (Okamoto et al, 1999).…”
