The efficacy and safety of calcitriol and/or Caltrate D in elderly Chinese women with low bone mass

Abstract: Aim:To observe the efficacy and safety of Rocaltrol (calcitriol) . The stand and maximal forward reach test (SMFRT) was significantly enhanced in both groups during the 12 months of treatment, but no significant differences were found between these two groups. No severe adverse event related to these medications occurred throughout the study. Conclusion: Treatment with Rocaltrol plus Caltrate D or Caltrate D for 12 months in elderly Chinese postmenopausal women effectively increased BMD at the lumbar spine. … Show more

“…In the OVX + HP group, not only was BMD increased, but the structure was also restored in the bones of the osteoporotic rats to a state that was comparable or improved compared to the OVX + MP and OVX + LP groups. In fact, some of the positive effects of phytoestrogen treatment that we observed have been previously reported (Fanti et al, 1998;Xia et al, 2009) in studies involving ovariectomized rats. Bone Gla protein (BGP), also referred to as osteocalcin, is the most plentiful noncollagenous protein of the bone matrix.…”

“…Calcitriol plays an important role in skeletal system regulation and may have other bone-protective effects independent of its calcium absorption effects [30] . To prevent hypercalcinuria, patients with osteoporosis or osteopenia in China are frequently treated with 0.25 μg/d of calcitriol [13] . After 3 months of intervention in our study, no differences were identified in calcium metabolism and bone turnover between the group treated with 0.25 µg of calcitriol daily and the group treated with 800 IU of cholecalciferol daily.…”

“…In elderly people, diminished renal function due to aging leads to a decrease in renal conversion to 1,25(OH) 2 D 3 [12] . Calcitriol [1,25(OH) 2 D 3 ] plays an important role in promoting bone formation, and D-hormone analogs prevent falls to a greater extent than does ordinary vitamin D. Therefore, the use of D-hormone analogs has been advocated for the treatment of osteoporosis in many studies [13][14][15] . Because patients who are treated with 0.5 μg/d or 1.0 μg/d of calcitriol tend to develop hypercalcinuria, serum and urinary calcium levels should be examined regularly, and the calcitriol regime should be examined intermittently.…”

“…It was later hypothesized that calcitriol plays a crucial role in skeletal system regulation due to its effect on calcium absorption and may have additional bone-protective effects independent of its calcium absorption effects [41, 42]. In addition to its positive effects on the skeletal system, calcitriol plays a vital role in muscular function in which higher levels are associated with increased muscular strength and balance [4345]. Although diminished BMD increases the risk of fracture, falls play an important role too [46, 47].…”
