The Effects of Xenobiotics on the Structure and Function of Thyroid Follicular and C-Cells

Abstract: The mammalian thyroid gland is composed of 2 distinct endocrine cell populations concerned with the synthesis of 2 different classes of hormones. Follicular cells secrete the metabolically active iodothyronines whereas the C-(parafollicular) cells are concerned with the production ofcalcitonin, a hormone that influences blood levels of calcium and phosphorus, and bone cell metabolism. The synthesis of metabolic thyroid hormones is different than in other endocrine glands because the final assembly of hormone o… Show more

“…Although perchlorate has been used as a treatment for hyperthyroidism, under the right circumstances it also can act as goitrogen in rodents and prevent thyroid hormone formation by interfering with iodide uptake (Capen and Martin, 1989). The low level of hormones is recognized by the pituitary gland which then stimulates the thyroid gland to work harder, eventually leading to goiter.…”

“…These presumably resulted from long-term direct effects of organochlorines on thyroid structure, including the deterioration of cell membranes (6,28), interference with peripheral thyroid hormone metabolism, immunomodulatory and other non-specific effects. These effects may have resulted in temporary or periodic variations in the effect of TSH.…”

“…Since these were distinct from those observed after short-and long-term thyroid-stimulating hormone (TSH) stimulation, they were considered to result from direct damage to the thyroid (6). Direct effects of PCBs on the thyroid are supported by the decreased effect of TSH on T 4 and triiodothyronine (T 3 ) release from the thyroid in rats pretreated with PCBs when compared with controls (7).…”

“…The secretion of thyroid hormones, under the influence of pituitary thyrotropin (TSH) from stores in the luminal colloid is initiated by elongation of microvilli and formation of pseudopods. D and C Red No.3 are tetraiodinated derivatives of fluorescein which in lifetime studies increases the incidence of thyroid follicular cell adenomas in male rats (Capen and Martin, 1989)). …”
