The Effects of Short-Term Immunosuppressive Therapy on Redox Parameters in the Livers of Pregnant Wistar Rats

Abstract: Immunosuppressive drugs are widely used to avoid graft rejection, but they are also known to be strongly hepatotoxic. The goal of the current study was to determine: (i) the immunoexpression of SOD1, CAT, GPX1; (ii) the concentration of MDA, GSH; (iii) the activity of SOD, CAT, GPX, in the native liver of a pregnant female rats undergoing immunosuppressive therapy. The study was based on archival material obtained from Department of Nephrology, Transplantology and Internal Medicine of the Independent Public Cl… Show more

“…It seems that immunosuppressive drugs affect blood xenobiotic levels, as was documented in our previous study on renal tissue [ 32 ], where we examined the levels of xenobiotic metals in the erythrocytes of renal transplant recipients, grouped by immunosuppressive regimen. The mechanism of action of immunosuppressive drugs is unclear, but it is known to be associated with oxidative stress [ 33 , 34 , 35 ]. Interestingly, in blood erythrocytes of renal transplant recipients from Szczecin, we observed significantly higher Pb level in patients treated with mycophenolate mofetil with tacrolimus than in patients who used mycophenolate mofetil together with cyclosporine A ( p = 0.002).…”

“…After preparation, the supernatants were stored at −80°C and used for the analysis of the parameters of oxidative stress. All biochemical analyses, including oxidative stress parameters, were performed according to manufacturer’s instruction, as described previously [3,20]. The product of the lipid peroxidation MDA was estimated using a Bioxytech LPO-586 Assay Kit (Oxis Research, Poland).…”

“…Little is known about the effect of IDs on hormonal status. However, histological and biochemical experiments conducted on male rats and pregnant female rats revealed that multidrug IT, in various regimens, significantly affected the liver and kidney [ 17 , 18 , 19 ]. There are data showing that CNIs inhibit the growth and steroidogenic capacity of Leydig cells, which are responsible for, among the other, T synthesis [ 6 , 20 ].…”
