Objectives: To challenge a vapour fast freezing (VFF) cryopreservation procedure (conventional VFF) with several vitrification protocols and VFF conducted with small semen volumes (10 µl, microVFF), in order to implement a procedure for sperm banking in subjects with small sperm number. Materials and methods:Conventional VFF was conducted with test yolk buffer (TYB) as freezing medium and 500 µl straws as carriers. MicroVFF was conducted with TYB and using tips or cell sleepers as carriers. Vitrification was performed with TYB or SpermFreeze as freezing medium and with microspheres and tips as carriers. The effect of different procedures on progressive and total motility, viability, oxidative stress and DNA fragmentation of spermatozoa (sDF) was determined. Fresh and thawed samples, the latter after adequate washing/centrifuging, were evaluated. In some experiments, motility and viability recovery was determined in thawed samples, omitting the washing/centrifuging step.Results: All the cryopreservation procedures blunted sperm motility and viability and induced increase of oxidative stress and sDF. However, VFF better preserved sperm motility and viability and less induced oxidative stress and sDF than vitrification, independently from the freezing medium and the carriers used in the latter. MicroVFF with cell sleepers resulted in a percentage increase of 57.58 ± 63.63%, 48.82 ± 74.96% and 24.55 ± 39.20% of, respectively, progressive and total motility and viability compared to the conventional VFF. Further, when tips were used, microVFF resulted in a percentage decrease of 15.77 ± 20.77% of sDF with respect to conventional VFF.Finally, omission of washing/centrifuging in post thawed samples, resulted in a much lower negative effect on motility and viability.Discussion and conclusion: VFF, and in particular microVFF, better prevents sperm cryodamage than vitrification. Washing/centrifuging step after sample thawing seems to be responsible for a relevant fraction of damage to sperm motility and viability. The cooling rate is a crucial factor in the process of freezing semen, influencing the overall freezing effectiveness. The height and time of fumigation can significantly impact the rate of cooling. Appropriate cooling rates can help minimize the formation of ice crystals in spermatozoa and reduce potential damage to them. Therefore, the aim of this study was to evaluate the effect of different fumigation heights and time for the cryopreservation of Hu ram semen. Experiments I–IV assessed the effect of semen cryopreservation by testing the post-thawed spermatozoa total motility (TM), progressive motility (PM) and kinetic parameters fumigated at distances of 2, 4, 6 and 8 cm for durations of 5, 10, 15 and 20 min, respectively. Based on the results of experiments I to IV, experiment V evaluated the effect of semen cryopreservation by testing the post-thawed spermatozoa TM, PM, kinetic parameters, plasma membrane integrity, acrosome integrity and reactive oxygen species (ROS) level fumigated at distances of 2, 4, 6 and 8 cm for duration of 20 min. The results indicated that fumigation at 2 cm for 20 min significantly (P < 0.05) improved spermatozoa TM, PM, mean angular displacement (MAD), plasma membrane integrity and acrosome integrity compared to other groups. Additionally, it significantly (P < 0.05) reduced spermatozoa ROS level compared to the 6 and 8 cm groups. In conclusion, fumigation for 20 min at a distance of 2 cm from the liquid nitrogen surface is the most suitable cooling method for the cryopreservation of Hu ram semen.

Our study aimed to enhance cryoprotection and improve post-thaw sperm quality by using a new freezing protocolthat combines trehalose and glycerol.The semen ejaculate from each bull was collected and split into three aliquots.Split one was the control group (C), in which semen was extended in a tris fructose egg yolk glycerol (TFEG) extender and underwent the conventional freezing protocol.In split two (T1) and split three (T2) groups, the semen was diluted in tris fructose egg yolk extender containing 25% trehalose, with glycerol 5% in both groups. Split two (T1) underwent conventional freezing, whereas split three (T2) underwent aluminium block freezing.After freezing-thawing, the progressive sperm motility,viability, acrosome integrity, sperm velocity and path parameters were significantly (p ≤ 0.05) improved in T2 among groups. Additionally, the degree of oxidative stress was substantially lower in T2 among groups.The present study's findings revealed the promising role of trehalose (25%) and glycerol (5%) in tris-based extender in achieving aluminium block freezing, as it significantly improved the post-thaw sperm survivability, and this method can suitably be used for quality frozen semen production.
