The first three essential amino acids (EAA) for broilers including methionine (Met), lysine (Lys) and threonine (Thr) may greatly influence the growth of chick muscles at early stages of life. In order to survey the potential effects of those EAA on growth muscles, a rotatable three-variable central composite design (CCD) was conducted to track the interrelationships of dietary digestible Met (dMet), Lys (dLys) and Thr (dThr) for optimization of processing yields in broiler chicks using response surface methodology. A total of 60 floor pens of six birds each were assigned to 15 dietary treatments based on CCD containing five levels of dMet (0.416% to 0.584% of diet), dLys (0.881% to 1.319% of diet) and dThr (0.532% to 0.868% of diet) from 3 to 16 days of age. Experimental treatments significantly affected breast mass (BM) and leg mass (LM) of the birds ( P < 0.05) in which the main effect of dLys on BM was threefold higher than the main effect of dThr, and interaction effect between dMet and dLys was observed on BM ( P < 0.05). However, in the case of LM, the main effect of dThr was higher than the main effects of dMet and dLys and highest interaction effect exist between dThr and dMet ( P < 0.05). The second-order models for BM and LM were fitted by least squares regression. Canonical analysis revealed that the stationary points for carcass components were saddle points, thus ridge analysis was performed for getting optimal values of each EAA. Ridge analyses of BM and LM models showed that the maximum BM point may be obtained with 0.58%, 1.05% and 0.76% of dMet, dLys and dThr, respectively, in diet, and maximum LM point may be achieved with 0.58%, 1.09% and 0.70% of dMet, dLys and dThr, respectively, in diet. The resultant ideal ratios of dMet and dThr to dLys were 55% and 72% for BM; 53% and 64% for LM. Moreover, sensitivity analysis showed that the most important amino acids in BM and LM models were Lys and Thr, respectively. In conclusion, providing these three amino acid for BM optimization may warrant LM optimization and higher ideal ratios of dMet and dThr for breast muscle may indicate the higher importance of these EAA in this muscle than those in thigh muscle.

ABSTRAKTujuan dari penelitian ini adalah mengetahui pengaruh pemberian pakan dengan sumber protein berbeda terhadap persentase potongan karkas dan massa protein daging ayam lokal persilangan. Materi yang digunakan adalah 126 ekor ayam lokal persilangan (unsexed) umur2 minggu dengan bobot badan 129 ± 6,34 g. Bahan pakan yang digunakan yaitu jagung, bekatul, bungkil kedelai, tepung ikan, MBM (Meat bone meal) dan PMM (Poultry meat meal). Rancangan percobaan yang digunakan adalah RAL (Rancangan acak lengkap) dengan 3 perlakuan dan 7 ulangan. Perlakuan yang diberikan yaitu T1 = 2 sumber protein (tepung ikan dan bungkil kedelai), T2 = 3 sumber protein (tepung ikan, bungkil kedelai, dan MBM), T3 = 4 sumber protein (tepung ikan, bungkil kedelai, MBM dan PMM). Pemberian pakan menggunakan metode bebas memilih dan perlakuan dimulai pada umur 2 minggu sampai 10 minggu. Parameter yang diamati adalah bobot akhir, bobot potongan karkas, persentase potongan karkas dan massa protein daging. Data yang diperoleh dianalisis ragam dengan uji F dengan taraf 5% dan pada penelitian ini terdapat pengaruh perlakuan sehingga dilanjutkan dengan uji jarak berganda duncan. Hasil penelitian menunjukkan bahwa pemberian pakan dengan sumber protein berbeda berpengaruh nyata (P<0,05) terhadap bobot akhir, bobot potongan karkas dan massa protein daging serta tidak berpengaruh nyata (P>0,05) terhadap persentase potongan karkas. Kesimpulan penelitian ini adalah pemberian pakan dengan sumber protein yang semakin beragam dapat meningkatkan bobot akhir, bobot potongan karkas dan massa protein daging serta memberikan hasil yang sama pada persentase potongan karkas ayam lokal persilangan.

RESUMO -Foram realizados dois experimentos para determinar a exigência nutricional de lisina total para frangos de corte Hubbard e Ross de ambos os sexos, no período de 22 a 40 e de 42 a 48 dias de idade. Foram avaliados ganho de peso, consumo de ração, conversão alimentary e rendimento de peito com osso e carne de peito. Para cada fase, o delineamento experimental utilizado foi o de blocos casualizados, em esquema fatorial 2 x 2 x 6, constituídos de dois sexos, duas marcas e seis níveis dietéticos de lisina, com quatro repetições e um total de 1920 aves. Dietas basais com 0,80% de lisina, 19,8% PB e 3100 kcal EM/kg, no primeiro experimento, e 0,75% de lisina, 18,5% PB e 3200 kcal EM/kg, no segundo experimento, foram suplementadas com seis níveis de lisina (0; 0,06; 0,12; 0,18; 0,24; e 0,30%). Considerando-se ganho de peso, conversão alimentar, rendimentos de peito com osso e carne de peito dos frangos, as estimativas das exigências dietéticas de lisina total na fase de 22 a 40 dias foram 0,80% para fêmeas Ross, 0,993% para fêmeas Hubbard, 0,998% para machos Hubbard e 1,03% para machos Ross, respectivamente. The basal diets with 0.80% of lysine, 19.8% CP and 3100 kcal ME/kg in the first experiment and 0.75% lysine, 18.5% CP and 3200 kcal ME/kg in the second experiment, were supplemented with six lysine levels (0, 0.06, 0.12, 0.18, 0.24 and 0.30%). Considering weight gain, feed:gain ratio, breast with bone yield and breast meat yield of the chickens, the total lysine dietary requirement estimates from 22 to 40 days were 0.80% for females Ross, 0.993% for female Hubbard, 0.998% for males Hubbard and 1.03% for males Ross, respectively.
