Woody cutting is customarily utilized as material in research of grape adventitious root formation (ARF).However, phenotypic heterogeneity caused by the complex background in uenced its use for molecular mechanism research of ARF of grape. The present study tested various types of explants from grape tissue culture plantlets and found the whole leaf: blade with petiole (LP) was the simplest unit that can easily form adventitious root (AR). LP explants which can be easily obtained, directly generate ARs via de novo organogenesis from the base of the petiole. The plantlet age, node position, blade size, the health condition of leaves, and light intensity have been demonstrated to affect the homogeneity of ARF phenotype in LP. By controlling these parameters, selected LPs cultured on medium with 6 g•L -1 agar and 10 g•L -1 sucrose under dark conditions started rooting at 6-7 day after culture (DAC) and reached 100% rooting rate within 13-14 DAC. Using this system, the core role of auxin on ARF was veri ed by exogenous application of indole butyric acid (IBA) and N-1-naphthylphthalamic acid (NPA). Strikingly, we found light promoted ARF in the absence of sucrose, but inhibited ARF in the presence of sucrose (10 g•L -1 ), while a low concentration of 0.1 mg•L -1 NPA partially relieved the inhibition. Finally, this study con rmed exogenous plant growth regulators (PGRs), including 6-benzyl aminopurine (6-BA), gibberellic acid 3 (GA 3 ), and 2,4-Epibrassinolide (EBR) inhibited ARF signi cantly. This simple, rapid, quanti able ARF research system provides a new approach to study the factors in uencing the formation and development of grape adventitious roots and establishes a framework for investigating the mechanism of grape adventitious root induction and initiation.

The conventional reproduction methods are not efficient for regeneration of Sumac (Rhus coriaria L.). The purpose of this work was to study the micropropagation of R. coriaria using lateral buds as explant in Murashige and Skoog (MS) medium with different concentrations of plant growth regulator (PGRs). Four concentrations of Benzylaminopurine (BAP) in combination with three concentrations of indol-3-butyric acid (IBA) and 1.0 mg/L gibberellic acid (GA3) were tested for establishment and shoot multiplication. For root induction, IBA was used at four levels combined with 0, 0.5 and 1 mg/L of naphthalene acetic acid (NAA) in full and half strength of MS medium. BAP at 2 mg/L with 1 mg/L IBA was best, with 88.88% of establishment. The highest shoot proliferation (12.30 ± 0.30) was obtained in medium fortified with 2 mg/L BAP plus 0.5 mg/L IBA and the highest shoot length (8.50 cm) was obtained at 3 mg/L BAP plus 1 mg/L IBA. The highest rooting (100%) was observed in 1/2-strength MS medium containing 1 mg/L IBA with 0.5 mg/L NAA. In conclusion, an efficient protocol with high rate of proliferation and rooting is described for R. coriaria, which can be used in massive propagation.

Toboali pineapple variety is a smooth cayenne variety because it contains high antioxidants in the form of vitamins A and C. In addition, it is a source of citric and malic acids, substances that can be used to enhance the taste of the fruit. Pineapple is a plant that is propagated with vegetative organs such as suckers, slips, and crowns. However, conventional propagation has a low propagation rate. Tissue culture is an in vitro technique used in the rapid propagation of plants and in large quantities to meet the needs of cultivation and research. The selection of regeneration media is very influential on the development of plants in tissue culture. This study aimed to obtain the optimal media combination with BAP and NAA in the regeneration of Smooth Cayenne pineapple (Toboali) in vitro for mass propagation of seedlings and genetic engineering research. This study used 6 levels of BAP, namely 0, 1, 2, 3, 4, 5 mg.L-1 and 4 levels of NAA, namely 0, 1, 2, 3 mg.L-1. The treatment used was a combination of BAP and NAA as many as 24 treatments with 3 replications. The best pineapple regeneration medium for mass plant propagation in this study was BAP 3 mg.L-1 without NAA media which produced the highest shoots of 17.33 shoots per explant at 12 week after planting. The best pineapple regeneration medium for genetic engineering research was a combination BAP 3 mg.L-1 and NAA 2 mg.L-1 media which produced 18.33 shoots per explant from embryogenic callus.
