The effects of chronic renal insufficiency on the pharmacokinetics of doxycycline in man.

Abstract: The pharmacokinetics as well as erythrocyte and plasma protein binding of doxycycline were studied in fifteen patients with various renal function impairments after oral doxycycline polyphosphate single administration. Plasma half‐life (t 1/2), area under the plasma concentration‐time curve (AUC), urinary excretion, renal clearance, erythrocyte and plasma protein binding (%) were regressed vs creatinine clearance. No significant correlations were observed between t 1/2 or AUC and renal function nor plasma prot… Show more

“…In this respect, it is centration curve 27 that falls below 1 g/ml after approximately 24 h for minocycline 30 and 50 h for doxycycline. 32 interesting to realise that anhydrotetracycline results in a shorter delay in resumption of tRP activity than minocycThis, together with the higher tissue to plasma concentration ratio of doxycycline and minocycline, ensures that line or doxycycline, despite having much higher K A for tetR compared with these derivatives. Moreover, tRP the expression of a given gene can be kept switched off …”

“…The t'/2 values of tetracycline, demeclocycline and oxytetracycline are prolonged in patients with renal dysfunction as they are primarily excreted unchanged in the urine (Saivin & Houin 1988). Although t'/2 values of doxycycline and minocycline may be slightly prolonged in renal failure, AVC and CL are not significantly altered Heaney & Eknoyan 1978;Houin et al 1983b;Sklenar et al 1977). Only Bernard and colleagues (1971) reported a greatly prolonged mean minocycline t,;, in patients with chronic renal failure when compared with healthy volunteers (30.3 vs 13.7h.…”
