The effects of chlorpromazine on psychomotor stimulant self-administration in the Rhesus monkey

Abstract: The effects of acute and chronic chlorpromazine treatment on psychomotor stimulant self-administration behavior in the Rhesus monkey were determined. Chlorpromazine treatment significantly increased the frequency of self-administration of cocaine, pipradrol, phenmetrazine, d-amphetamine and methyIphenidate. The basis of this effect was thought to either be due to an antagonism of the reinforcing effect of these compounds or an antagonism of those actions of the psychomotor stimulants which may function in limi… Show more

“…This type of comparison is not typically conducted for dopamine agonists. In fact, to control for "drug history" in cocaine substitution studies, many investigators include experimentally-naive subjects that are trained to self-administer cocaine at the start of the study (e.g., Wilson and Schuster 1972;Woolverton et al 1984;Lamb and Griffiths 1990;Katz et al 1992). It has been assumed that drugs that function as reinforcers in substitution paradigms have abuse liability, and this has certainly proven valid for the majority of compounds.…”

“…Dopaminergic antagonists and lesions of dopamine-containing cell bodies attenuate stimulant-induced psychomotor activation, self-administration, and c-fos expression (Schlechter and Butcher, 1972;Wilson and Schuster, 1972;Hollister et al, 1974;Roberts et al, 1977Roberts et al, , 1989Graybiel et al, 1990;Bhat and Baraban, 1993;Cenci and Bjorklund, 1994). Furthermore, HFS of the STN increases the therapeutic efficacy of L-DOPA in patients with Parkinson's disease.…”

“…Moreover, co caine's acute subjective effects and heart rate increases were greater in polydrug users on methadone main tenance (50 mg/day) than in abstinent opioid abusers (Preston et al 1993) . Preclinical studies of the effects of opioid agonists on cocaine self-administration by monkeys are also inconsistent; both no effect (Stretch 1977) and decreased cocaine self-administration have been reported (Wilson and Schuster 1970;Winger et al 1992) .…”
