Xanthone derivatives, a class of natural compounds abundantly found in plants such as mangosteen (Garcinia mangostana) and certain herbs, have garnered substantial interest due to their diverse pharmacological properties, including antioxidant, anti-inflammatory, and anti-cancer activities. Recent investigations have unveiled their potential as modulators of enzymatic activity, prompting exploration into their effects on hyaluronidase-mediated hyaluronic acid (HA) degradation, and their effects in topical treatment of telangiectasia and rosacea. Telangiectasia and rosacea are common dermatological conditions characterized by chronic skin inflammation, vascular abnormalities, and visible blood vessels, resulting in significant cosmetic concerns and impaired quality of life for affected individuals. This review aims to provide a comprehensive overview of the current understanding regarding the interplay between the mechanisms of action by which xanthone derivatives exert their therapeutic effects, including the inhibition of pro-inflammatory cytokines, modulation of oxidative stress pathways, and regulation of vascular endothelial growth factors. Furthermore, we will discuss the implications of harnessing xanthone derivatives as therapeutic agents for mitigating vascular dysfunction and its associated pathologies, thereby offering insights into future research directions and therapeutic strategies in the field of vascular biology.

Oxidative stress is implicated in the initiation, pathogenesis, and progression of various gastric inflammatory diseases (GID). The prevalence of these diseases remains a concern along with the increasing risks of adverse effects in current clinical interventions. Hence, new gastroprotective agents capable of inhibiting oxidative stress by modulating cellular defense systems such as the nuclear factor erythroid 2‐related factor 2 (Nrf2)/antioxidant response element (ARE) signaling pathway are critically needed to address these issues. A candidate to solve the present issue is xanthone, a natural compound that reportedly exerts gastroprotective effects via antioxidant, anti‐inflammatory, and cytoprotective mechanisms. Moreover, xanthone derivatives were shown to modulate the Nrf2/ARE signaling pathway to counter oxidative stress in both in vitro and in vivo models. Thirteen natural xanthones have demonstrated the ability to modulate the Nrf2/ARE signaling pathway and have high potential as lead compounds for GID as indicated by their in vivo gastroprotective action–particularly mangiferin (2), α‐mangostin (3), and γ‐mangostin (4). Further studies on these compounds are recommended to validate the Nrf2 modulatory ability in relation to their gastroprotective action.

Xanthones, a class of polyketide derivatives characterized by a dibenzo‐γ‐pyrone core structure, are aromatic oxygenated cyclic compounds extensively found in nature. They are known for their diverse biological activities, including anti‐Alzheimer, antitumor, and antidiabetic properties, which have spurred extensive research into their therapeutic potential. One of the most significant challenges facing mankind currently is the development of antibiotic resistance in pathogenic microbes making microbial infections increasingly difficult to treat. The formation of biofilms enhances bacterial resistance to antibiotics, immune defenses, and environmental stresses. In this review, we have categorized xanthones, both of biological origin and synthesized, based on their structure and summarized the recent studies on their ability to inhibit Gram‐positive and ‐negative bacteria as well as the various underlying mechanisms that contribute to their microbicidal activity. These mechanisms include inhibition of virulence factors and biofilm formation, interaction with cell membranes, and interference with cell wall synthesis and DNA replication. In addition, we have listed the various methods involved in biofilm inhibition and the structure‐activity relationship of various xanthone molecules, highlighting how xanthone derivatives exhibit increased antibacterial activity. With the growing concern of antibiotic resistance, exploring natural compounds like xanthones to study their interaction with bacterial cells is crucial for identifying new treatment options.
