The Effect of the Partial Obstruction Site of the Renal Vein on Testis and Kidney in Rats: Is the Traditional Animal Model Suitable for Varicocele Research?

Abstract: PurposeWe investigated the influence of the location of the partial renal vein obstruction on the left kidney, the bilateral testes, and cauda epididymal sperm quality and determined whether this animal model is suitable for varicocele study.Materials and MethodsA total of 25 adult male Sprague-Dawley rats were assigned to three groups: group 1 (experimental varicocele by partial ligation medial to the internal spermatic vein for 8 weeks, n=8), group 2 (partial ligation lateral to the internal spermatic vein f… Show more

“…Rats were divided into seven groups: two controls, two shams and three varicocele induction groups. Left‐side varicocele was induced according to previous studies (Ko et al, ; Razi, Sadrkhanloo, Malekinejad, & Sarafzadeh‐Rezaei, ), and two months after surgery, ten rats were sacrificed to verify our varicocele model by assessment of study parameters. From the 20 remaining rats in varicocele groups, 10 rats were treated with ALA [(±)‐α‐lipoic acid (T1395 Sigma)] by oral gavage (300 mg/kg of body weight) and the other 10 rats received equivalent amount of water for 2 months.…”

“…Samples of spermatozoa were collected from the caudal region of epididymis by mincing it finely in normal saline containing 0.5% bovine serum albumin at 37°C and then were filtered. Sperm suspensions were analyzed as previously described [ 14 ]. The sperm count represents the number of sperms in 1 mL of the medium.…”

“…The rats were randomly divided into 3 groups of 10 individuals. In the first group, the left varicocele was surgically induced (varicocele group) according to the protocol described in Ko et al [23]. In the second group, the rats underwent a sham laparotomy (sham group).…”
