“…This difference in sensitivity to TGF-␤ was confirmed by analysis of CFSE proliferation profiles of both CD4 ϩ and CD8 ϩ T cells in the naive (CD44 low ) T cell pool. In agreement with earlier in vitro studies using CD4 ϩ T cells, lymphocytes in the memory-effector pool (defined in this report as CD44 high ) were less susceptible to TGF-␤-induced growth arrest, regardless of genotype (data not shown) (18).…”

“…It has been reported previously that naive T cells are more sensitive to TGF-␤-mediated effects than are Ag-experienced ("memory" plus "effector") T lymphocytes (18). Thus we hypothesized that the apparent reduced sensitivity to TGF-␤ observed in cultures of DKO whole splenocytes might be due to the influence of Agexperienced T cells, particularly if their number or their potential for expansion in vitro is increased by the concomitant deletion of p21 Cip1 and p27 Kip1 .…”

“…It has been shown that TGF-b strongly inhibits Th1 cell differentiation from naive CD4 + T cells (37). Furthermore, Foxp3 in CD4 + T cells has been shown to suppress the production of various cytokines, including IFN-g. To clarify the role of Smad2 and Smad3 in TGF-b-mediated suppression of Th1 cell differentiation, and its relationship to Foxp3 induction, we compared Th1/iTreg differentiation among Smad2 …”

“…We currently are investigating several hypotheses on suppressive mechanism and imprinting of Th2-type phenotype on naive T cells in coculture. Although we were able to show that soluble factors such as IL-10, IFN-␥, and NO were involved in suppression, we were surprised to find that blockade of TGF-␤, a cytokine known to induce suppression in activated T cells (31,38) did not restore CD4 ϩ proliferation. However, the suppressive effect of TGF-␤ is not restricted to inhibition of proliferative responses, but in addition, it has the ability to modulate the profile of activating cytokines, such as limiting the production of IFN-␥ (31, 38).…”
