The Effect of Sodium Pump Blockade and Denervation on the Steady‐state Sodium Permeability of Mouse Skeletal Muscle Fibres

Abstract: SUMMARYThe Na+ pump inhibitor ouabain produced a 25 mV depolarization of mouse extensor digitorum longus muscle fibres in 2 h. Approximately 40 % of this depolarization could be prevented by the inclusion of tetrodotoxin (TTX) in the incubation fluid, and the membrane potential of ouabain-depolarized fibres increased when external Na+ ion concentration was reduced. TTX and Na+ reduction were without effect on fibres incubated for 2 h in the control saline or on fibres depolarized by raising extemal K' ion conc… Show more

“…In the sartorius muscle of the frog and the diaphragm muscle of the rat, however, K+ can be reduced to at least 0.1 mm (Hodgkin & Horowicz, 1959) or even to ('nominally') potassium-free media (Hinke, 1987;Kuba & Nohmi, 1987) without depolarization. Other skeletal muscle cells do show the expected dichotomy in recorded Vm values when K+ is reduced (M0lgaard, Stiirup-Johansen & Flatman, 1980;Seabrooke, Ward & White, 1988;this paper). The present study shows that the dichotomy can be measured in a single cell.…”

“…Since barium blocks both Na + and K + channels, we perfused 8 hindlimbs with the voltage-sensitive Na + channel blocker TTX (Seabrooke et al 1988;Santana et al 1998) to discriminate between K + efflux through K + channels (most likely candidate) versus voltage-sensitive Na + channels. This was followed by 20 min of perfusion with TTX and 5 mM ouabain combined to determine if TTX-sensitive Na + chan-nels are involved in the ouabain-induced increase in unidirectional K + efflux.…”

“…In vitro, peak depolarizing effects (5-20 mV) have been observed with 1 mM ouabain (Clausen and Flatman 1977;Rogus et al 1977;Seabrooke et al 1988). Reductions in membrane potential may in turn affect voltage dependent ion channels (Seabrooke et al 1988) and other cellular proteins.…”
