The Effect of Sildenafil on Recuperation from Sciatic Nerve Injury in Rats

Abstract: Background: Severe functional and anatomical defects can be detected after the peripheral nerve injury. Pharmacological approaches are preferred rather than surgical treatment in the treatment of nerve injuries. Aims: The aim of this study is to perform histopathological, functional and bone densitometry examinations of the effects of sildenafil on nerve regeneration in a rat model of peripheral nerve crush injury. Study Design: Animal experiment. Methods: The study included a total of thirty adult Sprague-Daw… Show more

“…Additionally, inflammation of the central and peripheral nervous system helps to cause and sustain chronic neuropathic pain [1]. Issues related to crushing injuries include increased endoneurial edema, free oxygen radicals, inflammatory reactions, de-and remyelination, axonolysis, regeneration, and degeneration [2][3][4]. Inflammatory mediators such as prostaglandins, histamines, cytokines, chemokines, and growth factors are involved in nerve regeneration and degeneration that is associated with neuropathic pain [5][6][7].…”

“…2-6th weeks post-surgery, there were no significant differences between the groups in SFI. [ 141 ] Rat Sciatic nerve 17-beta-estradiol "Estrogen hormone " Transaction of about 3 mm 10 μL a of 17- beta-estradiol (0.1 mg/mL) Silicon conduit filled with estradiol was inserted at the lesion site by epineural suture Static Sciatic Index (SSI), Behavioral Testing, Electrophysiological Assessment, Biomechanical Testing, Muscle Mass and Histological Preparation and Morphometric Studies 1-At 12 weeks, the SFI estradiol group was higher than in the silicone group. 2-The muscle weight ratio in the Estradiol group was higher than in the silicon group.…”
