Molecular weights of alginates hydrolyzed with organic acids ranged from 7.5 to 53.2 kDa. There was no significant difference in the molar ratio of mannuronate to guluronate in alginates hydrolyzed with organic acids. Acetic acid was found to be the most effective organic acid for hydrolysis of alginate.The MW of alginate decreased with increasing concentration and reaction time with acetic acid as a hydrolyzing agent. The correlations between the MW of hydrolyzed alginate and concentration of acetic acid as well as reaction time with 0.4 M acetic acid were plotted and the relevant equations obtained in this study. Polymannuronate and polyguluronate were isolated by pH adjustment of alginate hydrolyzed with 0.4 M acetic acid. The molar percentages of mannuronate in polymannuronates isolated from alginate hydrolyzed with 0.4 M acetic acid at 100 o C were increasing in proportional to the reaction time such as 75% for 1 hr, 90% for 3 hr, and 98% for 5 hr of reaction time.

Effect of Sargassum confusum extract on the reduction of body weight gain and lipid contents in obese rats were evaluated to find natural materials with anti-obesity benefits. After inducing obesity by feeding 42.5% high-fat diet for 5 weeks, each 10 Sprague-Dawley rats were randomly assigned to high-fat diet control (HFD) group and high-fat diet group containing 3% Sargassum confusum extract (HFDSC). Weight gain of HFD group (2.96±0.31 g/day) was significantly (p<0.05) higher as compared to that of normal diet (ND) group (2.19±0.17 g/day). Weights of adipose tissues of HFD group were higher than those of ND group. Body weight gain of HFDSC group, however, was 2.36±0.24 g/day, which was significantly (p<0.05) lower by 21% than that of HFD group. In addition, weights of epididymal and perirenal adipose tissues were lower by 15% and 16%, respectively, as compared to those of HFD group. Biochemical analyses showed that concentration of triglyceride, total cholesterol, and fatty acids were significantly (p<0.05) lower in HFDSC group. These results suggest that Sargassum confusum extract has a high potential as an anti-obesity material by reducing weight gain and obesity-related factors in serum.

This study evaluated the anti-obesity effects of lactic acid bacteria. Thirty-one lactic acid bacteria were examined in vitro for their ability to inhibit α-glucosidase activity, lipase activity, and 3T3-L1 cell differentiation. Four selected lactic acid bacteria were administered to obese C57BL/6J mice models for 8 weeks. The degree of improvement in obesity was determined by weight gain and serum biochemical analysis. The expression levels of genes (Fas and Cpt-2) related to obesity in the liver were analyzed by quantitative reverse transcription (qRT)-PCR. In addition, antioxidant protein levels (SOD-2, CAT, and GPx-1) in the liver were evaluated. The lactic acid bacteria-treated groups (PPGK1, LFNK3, LPNK2, and LFNK4) showed lower weight increase rate than the control group. The total cholesterol (T-chol), triglyceride (TG), alanine aminotransferase (ALT), and aspartate aminotransferase (AST) levels in the blood serum of the LFNK4 group were the lowest among other groups, compared to the control group. The expression levels of lipid metabolism-related genes (Fas and Cpt-2) in the liver of the LFNK4 group were lower in Fas and higher in Cpt-2 than in the control group. The antioxidant protein expression levels (SOD-2, CAT, and GPx-1) in the liver tissue were also higher in the LFNK4 group. These results indicate that L. fermentum SMFM2017-NK4 has anti-obesity effects.
