The effect of non-traditional cooling on dog sperm cryosurvival and ability to perform the acrosome reaction

Abstract: The objective of this study was to assess cryosurvival, plasma membrane fluidity, and capability of cryopreserved dog (Canis lupus familiaris) spermatozoa, cooled to -5 °C before freezing, to perform the acrosome reaction under the effect of progesterone and calcium ionophore. In the first experiment, fresh spermatozoa diluted in Tyrode's medium plus albumin, lactate, and pyruvate (TALP) were incubated at 38 °C in 5% CO 2 in air, with progesterone or calcium ionophore added at 2, 4, and 6 h after incubation an… Show more

“…Our results on membrane fluidity support our hypothesis that high doses (0.002 and 0.0035 mol/L) of MLT significantly reduce the percentage of sperm with hyper-fluid membranes after freeze-thawing. However, similar results were obtained in dog sperm by the mere effect of prefreeze cooling (Ortega-Morales et al 2019). We argue that because the difference in the percentage of hyper-fluid membranes due to MLT is larger than that of cooling to -5 °C alone, there is an additive effect of MLT and cooling.…”

“…In a similar study, using a different breed of dogs (Belgian and German Shepherds), cooling reduced the percentage of hyper-fluid membranes in comparison to the usual cooling to +5 °C (Ortega-Morales et al 2019). These observations support the proposed hypothesis that the cooling of spermatozoa to subzero temperatures favours sperm plasma membrane reorganisation after the lipid phase transition takes place, thus avoiding an excessive increase in membrane fluidity (Watson 1995;Holt 2000).…”

“…Furthermore, the reduced of temperature during chilling processing which preserved at 5 °C or less inflicts sub-lethal damage to spermatozoa, compromising sperm quality and the success of artificial breeding as described by previous studies (Mohamed et al, 2020 andBustani andBaiee 2021). The earlier studies have concluded effect of chilling temperature on the sperm motility in both general and progressive motile of rat (Varisli et al, 2009), bull (Januskauskas et al, 1999), rabbit (Rosato et al, 2011), human (Verheyen et al, 1993) and, ram (Kulaksiz et al, 2012) which illustrated the temperature related in the organization of structural components of the plasma membrane have profound effects upon its properties, associated with changes in permeability, changes in membrane fluidity (Ortega et al, 2019), dis-regulation of intracellular Ca+' influx (Yeste et al, 2018) and changes in enzyme activity (Palhares et al, 2021) finally on the ability of sperm capacitation. From the Figure (1 and 2) the result showed the improved effect of Prunus cerasus (G1) on the sperm motility (general and progressive).…”

“…Notou-se, ainda na Tabela 3, que o grupo do ácido palmítico (G2) apresentou a melhor porcentagem de espermatozoides com MP íntegra (45,60%) se sobressaindo do G3 (39,80%), que também foi melhor que o controle (28,4%). A composição lipídica da membrana plasmática dos espermatozoides está diretamente relacionada à MOT e à fluidez dessa membrana [22].…”
