The effect of mepyramine and ranitidine on the oestrogen and anti‐oestrogen stimulated rat uterus

Abstract: 1 The aims of this study were to elucidate further the role of histamine in the rat uterotrophic response and to investigate the differences between the oestrogen and the anti-oestrogen induced uterine responses. 2 The parameters examined were uterine blood flow (measured by the microsphere technique), uterine wet and dry weights.3 17P-Oestradiol and the anti-oestrogen, tamoxifen, were used to stimulate the ovariectomised rat uterus and the antihistamines mepyramine (HI) and ranitidine (H2) were employed to mo… Show more

“…The classical effects of 17βE # on reproductive function [25,34,35], haemostasis and uterine smooth muscle contraction [36,37] are mediated via the nuclear oestrogen receptor [38,39]. These effects take some hours to develop, and are inhibited by protein synthesis inhibitors [25,40].…”

“…Depending on the species, histamine causes stimulation or inhibition of spontaneous contractions of myome trium acting on H r or H2-receptors, respectively. In rats, histamine relaxes by acting on H2-receptors located either on muscular myométrial cells or on adrenergic ter minals [1,2], In the human uterus, the action of hista mine is stimulation apparently associated with an inter action with H preceptors [3].…”

“…The agonist effect of tamoxifen on rat uterus has been demonstrated in this laboratory (Marshall & Senior, 1987), as have differences between the oestradiol-and tamoxifen-evoked responses (Marshall & Senior, 1986;1989). In these experiments the thromboxane antagonists did not significantly alter the tamoxifen response.…”
