The effect of melatonin on 7,12‐dimethyl‐benz[a]anthracene injury in comparison with vitamin E + selenium in mouse kidneys

Abstract: The aims of this study were to observe the changes in antioxidative defense enzymes and renal morphology after 7,12-dimethyl-benz[a]anthracene (7,12-DMBA) administration in mice and to investigate the possible protective effects of melatonin against 7,12-DMBA-induced renal damage in comparison with vitamin E + selenium (vit E + Se). Forty female mice were divided into four groups: control, DMBA, DMBA + vit E + Se and DMBA + melatonin. In the DMBA group, mice were given injections of 7,12-DMBA (20 mg/kg). DMBA … Show more

“…It is an essential cofactor in the hydroxylation of procollagen chains during collagen synthesis ( 21 ) . Vitamin E and Se also have antioxidant properties ( 19 , 22 , 23 ) . We fed rats with 10 % and 20 % apricot-containing feed for 5 months before CCl 4 exposure.…”

“…Considering the much lower molar concentration of MLT compared with vitamin E, MLT was a more potent antioxidant especially in the brain and kidney [31]. Likewise, it was reported that MLT has more protective role as an antioxidant in 7,12-dimethyl-benz[a]anthracene-induced injury in rat kidney and brain in comparison with VE plus Se [32,33]. Several another comparative studies have shown that VE plus Se prevents cadmium-induced oxidative damage in rat blood, kidney, testis, and liver more than MLT [34][35][36].…”

“…In vivo, it is an effective pharmacological agent in reducing oxidative damage under conditions in which excessive free radical generation is believed to be involved [13,14,19] The ability of MLT to reduce LPO may stem from its function as a preventive antioxidant (scavenging the initiating radicals) or from as yet unidentified actions. Moreover, peroxynitrite has also been shown to be directly scavenged by MLT [31][32][33]. Melatonin stimulates the activity or gene expression of several important endogenous antioxidant enzymes, including GSH-Px, SOD, and CAT.…”

“…Vitamin C is an excellent electron donor to free radicals which subsequently quench their deleterious activity on cellular macromolecules, thus playing a role in antioxidant mechanism (Sabiu et al, 2015). Also, vitamin E and Se also have antioxidant properties (Ozturk et al, 2006). Furthermore, β-carotene treatment along with APAP caused a significant reduction of APAP-induced elevation of AST, ALT, and ALP and appears to be protective in reducing the injurious effect of APAP (Morakinyo et al, 2012).…”
