The present study was planned to investigate the protective effect of 10 % and 20 % apricot-containing feed on carbon tetrachloride (CCl 4 )-induced hepatic steatosis and damage. Adult male Wistar rats (n 42) were divided into six groups of seven each, as follows: control group; CCl 4 group; CCl 4 þ 10 % apricot group; CCl 4 þ 20 % apricot group; 10 % apricot group; 20 % apricot group. All apricot groups were fed with 10 % or 20 % apricot-containing feed for 5 months. CCl 4 injections were applied to the CCl 4 groups at the dose of 1 mg/kg for 3 d at the end of 5 months. In the CCl 4 group, vacuolated hepatocytes and hepatic necrosis were seen, especially in the centrilobular area. Hepatocytes showed an oedematous cytoplasmic matrix, large lipid globules and degenerated organelles. The area of liver injury was found significantly decreased with apricot feeding. Malondialdehyde and total glutathione levels and catalase, superoxide dismutase and glutathione peroxidase activities were significantly changed in the CCl 4 group and indicated increased oxidative stress. Apricot feeding decreased this oxidative stress and ameliorated histological damage. We concluded that apricot feeding had beneficial effects on CCl 4 -induced liver steatosis and damage probably due to its antioxidant nutrient (b-carotene and vitamin) contents and high radical-scavenging capacity. Dietary intake of apricot can reduce the risk of liver steatosis and damage caused by free radicals.

Oxidative stress is considered to be the main cause of diabetic complications. In the current study, we investigated the effect of selenium-vitamin E combination and melatonin on lipid peroxidation (LPO) and scavenging enzyme activity in the blood of streptozocin (STZ)-induced diabetic pregnant rats. Forty female Wistar rats were randomly divided into five groups. The first and second groups were used as the non-pregnant control and pregnant control groups, respectively. The third group was the pregnant diabetic group. Vitamin E plus selenium and melatonin were administered to the diabetic pregnant rats consisting fourth and fifth groups, respectively. Diabetes was induced on day 0 of the study by STZ. Blood samples were taken from all animals on the 20th day of pregnancy. LPO level was higher in diabetic pregnant rats than in control, although superoxide dismutase, catalase, and glutathione peroxidase activities were lower in diabetic pregnant animals than in control. LPO levels were lower both in the two treatment groups than in the diabetic pregnant rats, whereas selenium-vitamin E combination and melatonin caused a significant increase in the activities of these antioxidant enzymes (p<0.01). In conclusion, vitamin E plus selenium seems to be a more potent antioxidant compared to melatonin in diabetic pregnant rats. Melatonin did not significantly affect the elevated glucose concentration of diabetic pregnant treated with melatonin group. Vitamin E plus selenium may play a role in preventing diabetes-related diseases of pregnant subjects.

This study was designed to investigate the protective effect of fresh, dried apricot and its kernel added to diet (10% and 20% w\w) on paracetamol (N-acetyl-p-aminophenol , APAP)-induced cellular toxicity in rats. Eighty male albino rats were used in this study, divided into 8 groups 10 rats each. Gp (1): set as healthy control, Gp (2) set as APAPaffected control. Gps (3-4) fed on diet supplemented with fresh apricot (10% & 20% w\w), Gps (5-6) fed on diet supplemented with dried apricot (10% & 20% w\w). Gps (7-8) fed on diet supplemented with apricot kernel (10% & 20% w\w). After 6 weeks, rats from groups (2:8) were injected with a single dose, i.p injection, of APAP (500 mg\kg body weight). After 24 h of injection, rats were sacrificed under diethyl ether anesthesia. Serum and tissue samples were collected to perform the biochemical analysis. Serum liver function tests [total protein, albumin, total bilirubin, direct and indirect bilirubin, the activities of alanine amino transferase (ALT), asparatet amino transferase (AST) and alkaline phosphatase (ALP)] were measured. Moreover, serum kidney function tests (urea, creatinine, and uric acid), and serum lipids profile were measured. Also, antioxidant status was evaluated by measuring reduced glutathione (GSH) and malondialdehyde (MDA) in liver and kidney homogenate. The results were supported with microscopic examination of liver and kidney tissues. The results of this study revealed that administration of APAP caused marked reduction in serum total protein, albumin and tissue GSH, in association with elevation of serum bilirubin, activities of AST, ALT, ALP, urea, creatinine, uric acid, lipids profile, and tissue MDA. On the other hand, supplementation of diet with fresh, dried apricots and apricot kernel had protective effect on liver and kidney tissues that was manifested by improving liver and kidney functions, antioxidant status, and lipids profile. The microscopic examination of liver and kidney tissues showed also amelioration in the appearance of cells. The protective effect of apricot and its kernel is due to their antioxidant potential that is represented by their rich content of dietary fibers, β-carotene, vitamin C, tocopherols, flavonoids, poly unsaturated fatty acids, and some polyphenols that appears to protect against reactive oxygen species (ROS) and consequently APAP toxicity.
