The effect of inhibition of endoplasmic reticulum stress on lipolysis in white adipose tissue in a rat model of chronic kidney disease

Abstract: Aim: Lipolysis in fat tissue plays an important role in the development of metabolic disturbances, a characteristic feature of chronic kidney disease (CKD). In the present study, we tested the hypothesis that the inhibition of endoplasmic reticulum (ER) stress could alleviate lipolysis in white adipose tissue in a rat model of CKD. Methods: A rat model of CKD was established by a method of reduced renal mass (RRM). Lipolysis was measured as the release of glycerol in ex vivo fat pads and cultured primary adipo… Show more

“…Our finding is in accordance with previous studies showing that hypoxia exposure induce ER stress in 3 T3-F442A and 3 T3-L1 adipocytes (Mihai & Schroder, 2015). UPR pathways were activated to ameliorate the overload of unfolded proteins under ER stress, which in turn influence lipid metabolism alleviated lipolysis in a rat model of chronic kidney disease (Zhu et al, 2014). In addition, curcumin was reported to suppress the ER stress-mediated lipolysis via cAMP/PKA/HSL pathway (Wang et al, 2016).…”

“…One explanation could be that the short complex lipids may be composed of relatively shorter FFAs (,12 carbons), which are less dependent on carnitine for mitochondrial transport. 50 Therefore, they become a preferred substrate for catabolic processes 42 and along with accelerated lipolysis from uremia, [51][52][53] contribute to the lower abundance of short saturated complex lipids in stage 5 CKD.…”

“…Secreted proteins play critical roles in physiological and pathological processes and can be used as biomarkers and therapies for aging and disease 1 , 2 , 3 , 4 , 5 , 6 , 7 . Because adipose tissue is the largest endocrine organ, we have focused on adipose-derived secreted proteins (also known as adipokines) and explored their functions over the past decade 8 , 9 , 10 , 11 , 12 , 13 , 14 , 15 , 16 .…”
