The effect of IHF on σ S selectivity of the phoA and pst promoters of Escherichia coli

Abstract: The pst operon, a member of the PHO regulon of Escherichia coli, encodes a high-affinity phosphate transport system whose expression is induced when the cells enter a phase of phosphate starvation. The expression of pst is stimulated by the integration host factor (IHF). Transcription of the PHO regulon genes is initiated by the RNA polymerase complexed with sigma (D) (Esigma (D)). Owing to a cytosine residue at position -13 of the pst promoter its transcription can also be initiated by Esigma (S). Here, we sh… Show more

“…As expected, the cultures in low-Pi medium displayed highly induced AP activity, confirming that they have entered the Pi-starvation phase while those grown in high-Pi showed only basal AP levels. PpstS showed the strongest CAT activity and a Pi-dependent induction of *11-fold (Table 2, line A), confirming previous data (Taschner et al 2006). Both the PpstC and PpstB clones presented a weaker but constitutive CAT activity (Table 2, lines B and D) and PpstA (line C) showed a negligible activity, lower than the negative control (line F).…”

“…The new plasmid constructs and pBS11 (pstS promoter cloned in pKK232-8) (Taschner et al 2006) were transformed into the wild-type strain MG1655 and into its isogenic phoB mutant (strain EY1491) to test if the cloned fragments display promoter activity and if they are phoBdependent. Each transformant was tested for growth on LB plates supplemented with chloramphenicol.…”

“…In bacteria, metabolism changes that allow cells to survive occur during environmental stress conditions (Weber et al, 2005;Jain et al, 2006;Klauck et al, 2007). Many lines of evidence suggest that the Pho regulon and the stress response are interrelated (Spira et al, 1995;Spira & Yagil, 1998;Ruiz & Silhavy, 2003;Taschner et al, 2004Taschner et al, , 2006Schurdell et al, 2007). Figure 2 illustrates interconnections between the Pho regulon and some key factors involved in nutritional and other stress responses.…”
