The Effect of TRANSPARENT TESTA2 on Seed Fatty Acid Biosynthesis and Tolerance to Environmental Stresses during Young Seedling Establishment in Arabidopsis

Abstract: In plants, fatty acids (FAs) and FA-derived complex lipids are major carbon and energy reserves in seeds. They are essential components of cellular membranes and cellular signal or hormone molecules. Although TRANSPARENT TESTA2 (TT2) is well studied for its function in regulating proanthocyanidin biosynthesis in the seed coat, little attention has been given to its role in affecting seed FA accumulation and tolerance to environmental stresses. We demonstrate that the tt2 mutation remarkably increased the seed … Show more

“…Seed FAs were extracted and analyzed as reported previously in detail (Poirier et al, 1999;Chen et al, 2012). Briefly, total FAs were converted to FA methyl esters in methanol solution containing 1 M HCl for 2 h at 80°C.…”

“…The negative feedback effect of TTG1 on LEC2 and ABI3 plays a role in mediating FUS3 expression in seeds, thereby regulating the rate of seed storage reserve accumulation (Chen et al, 2015). An R2R3-MYB protein, MYB123 (TT2), was found to be a key regulator of not only proanthocyanidin deposition but also oil accumulation in seeds (Nesi et al, 2001;Chen et al, 2012;Wang et al, 2014). Knockout of MYB118, which also is a member of the R2R3-MYB TFs, caused the derepression of maturation-related genes, thus stimulating storage processes in endosperm (Barthole et al, 2014).…”

“…Measurement of fatty acids in seeds was subsequently performed using the gas chromatograph (GC-2014, Shimadzu) as previously described (Poirier et al, 1999;Chen et al, 2012a). Starch and protein in seeds were measured as described before (Chen et al, 2012b).…”

“…In plants, flavonoid derivatives determine the pigmentation pattern of vegetative organs and seeds (Nesi et al, 2001; Lepiniec et al, 2006) and are involved in multiple functions. For example, they play important roles in protecting plants against a wide range of environmental injuries Agati and Tattini, 2010;Agati et al, 2012;Chen et al, 2012b). Seed coat mutants include two major groups.…”
