The coconut water effect combined with 5% of glycerol for preserving goramy spermatozoa at -34 °C for 48 hours has been studied. The objective of study is to find the best combination among 0%, 21%, 23%, 25% 27%, and 29%, respectively, of coconut water combined with 5% of glycerol for maintaining the good spermatozoa motility and viability, and minimizing spermatozoa abnormality. One part of semen/sperm were mixed with three parts of solvent (5% of glycerol + fish ringer + coconut water), and were equilibrated at 4 °C for 45 min. The diluted sperm were then freezed at -34 °C for 48 h. Cryopreserved sperms were thawed at 30 °C for 3-5 min. Spermatozoa quality were evaluated before and after sub-zero freezing. Based on Kruskal-Wallis test, spermatozoa motility and viability were higher than control (P<0.05), while the spermatozoa abnormality were not significantly different compared to control (P>0.05). Twenty five percent of coconut water combined with 5% of glycerol were the best combination for preserving spermatozoa motility (80.36±1.54)% and spermatozoa viability (82±1.86)%, and also minimized spermatozoa abnormality (10±1.03)%.

Sperm cryopreservation is one of the methods in preserving germplasm and avoid the extinction of native fish. The success of this technique requires proper use of protocol because each species needs a different response to certain protocols. However, very limited of works have been done in Indonesian fish; Presently, there were only 9 species of freshwater, namely; Channa striata, Chromobotia macracanthus, Barbonymus gonionotus, Mystus nemurus, Osphronemus goramy, Osteochilus hasseltii atau Osteochilus vittatus, Poropontius tawarensis, Rasbora tawarensis, and Tor soro, and one species marine fish Ephinephelus lanceolatus have been successfully developed the cryopreservation protocols. The objective of this review paper is to summarized and evaluate the best protocol for sperm cryopreservation of above species The reviewed aspects are including the type of extender and cryoprotectant, freezing process (storage time and temperature), thawing (thawing time and temperature) and observations (including the percentage of motility, viability, abnormality, fertility, and hatching rate). Oleh karena itu, penelitian ini mencoba merangkum dan mengevaluasi protokol terbaik dalam proses kriopreservasi masing-masing spesies yang tersebut diatas. Aspek yang direview adalah jenis extender, krioprotektan, proses freezing (lama penyimpanan sperma dan suhu), thawing (lama thawing dan suhu) dan data hasil pengamatan (meliputi persentase motilitas, viabilitas, abnormalitas, fertilitas dan hatching rate.Kata kunci: Extender, Krioprotektan, Freezing, Thawing

Sex determination and differentiation are processes by which a bipotential gonad adopts either a testicular or ovarian cell fate, and secondary sexual characteristics adopt either male or female developmental patterns. In birds, although genetic factors control the sex determination program, sex differentiation is sensitive to hormones, which can induce sex reversal when disturbed. Although these sex-reversed birds can form phenotypes opposite to their genotypes, none can experience complete sex reversal or produce offspring under natural conditions. Promising evidence indicates that the incomplete sex reversal is associated with cell autonomous sex identity (CASI) of avian cells, which is controlled by genetic factors. However, studies cannot clearly describe the regulatory mechanism of avian CASI and sex development at present, and these factors require further exploration. In spite of this, the abundant findings of avian sex research have provided theoretical bases for the progress of gender control technologies, which are being improved through interdisciplinary co-operation and will ultimately be employed in poultry production. In this review, we provide an overview of avian sex determination and differentiation and comprehensively summarize the research progress on sex reversal in birds, especially chickens. Importantly, we describe key issues faced by applying gender control systems in poultry production and chronologically summarize the development of avian sex control methods. In conclusion, this review provides unique perspectives for avian sex studies and helps scientists develop more advanced systems for sex regulation in birds.
