Sperm Response to in vitro Stress Conditions in Wild and Domestic Species Measured by Functional Variables and ROS Production

The domestication process has resulted in profound changes in the reproductive physiology of the animals that might have affected the sperm characteristics and thus their sensitivity to handling and cryopreservation procedures. This work assesses the response of the sperm of domestic and wild ungulates to a cooling storage at 15°C for 20 h followed by incubation at 38.5°C, 5% CO2, for 2 h. In addition, this paper examines the most representative sperm traits to assess their responsiveness to these stress conditions. Sperm samples were collected from domestic and their wild ancestor species: ram, mouflon, buck, Iberian ibex, domestic boar, and wild boar. Sperm motility, viability, mitochondrial membrane status, DNA fragmentation, and reactive oxygen species production were evaluated at the beginning of the experiment, after 20 h of refrigeration at 15°C, and, finally, at 2 h of incubation at 38.5°C. Sperm from all domestic species (ram, buck, and domestic boar) suffered more stress than their wild relatives (mouflon, Iberian Ibex, and wild boar). In pigs, the percentage of intact mitochondria was lower in the domestic species compared to wild boar. In sheep, we found a higher reactive oxygen species production in rams, while in goats, the curvilinear velocity was lower in the domestic species. The PCA (principal components analysis) showed that the motility and their kinetic variables were the most represented variables in the principal components of all species, indicating that they are essential biomarkers for evaluating the stress response. Sperm viability was highlighted as a representative variable for evaluating the stress response in domestic boar, mouflon, ram, and ibex.

Introducción. El uso de biotecnologías reproductivas ha significado una mejora relevante sobre los parámetros de eficiencia y rentabilidad en la industria porcina. Objetivo. Determinar el estado del arte en el que se encuentra la biotecnología aplicada al estudio del semen y la implementación; en granjas porcinas; de técnicas de reproducción asistida como la inseminación artificial. Desarrollo. En el sector porcino, la fertilidad de los verracos (Sus scrofa domestica) tiene un efecto significativo sobre la eficiencia general de la reproducción de las piaras. Aunque hay métodos objetivos de valoración precisa de las variables seminales de los verracos, muchas granjas aún continúan realizando sus evaluaciones de forma subjetiva. Los conceptos bioquímicos que determinan la funcionalidad del gameto masculino o espermatozoide son molecularmente complejos, sin embargo, pueden conocerse los aspectos generales de la funcionalidad y desarrollar métodos de manejo del semen que permita el mantenimiento de las condiciones óptimas de las muestras seminales o dosis de semen utilizadas en inseminación artificial (IA). Dentro de la funcionalidad espermática intervienen parámetros clásicos de volumen, concentración y movilidad que son fundamentales para la optimización de las dosis seminales producidas por verraco. Los sistemas CASA (Computerassisted semen analysis) son una aplicación biotecnológica que desplaza el análisis subjetivo tradicional del semen para la valoración de la calidad, estos aportan grandes volúmenes de información que han permitido explorar más en detalle los eyaculados porcinos. Conclusiones. En las granjas que utilizan la inseminación artificial, el uso de técnicas biotecnológicas para el análisis seminal, constituye un valor agregado para el estudio de la calidad del semen y la optimización de dosis seminales producidas.

In the United States, the Clinical Laboratory Improvement Act (CLIA) of 1988 describes requirements and guidelines for implementing a quality control/quality assurance (QC/QA) program for moderate and high complexity laboratories. These requirements and guidelines apply to Assisted Reproductive Technology (ART) laboratories as well. The general topic of QC and QA as it pertains to in vitro fertilization (IVF) and embryo transfer (ET) is extensively reviewed. This review summarizes many of the QC and QA events that contribute to the advancement of knowledge in this biotechnological field. These events include control of the culture environment inside and outside of the incubator, as well as factors that affect culture media. This review also discusses, in considerable detail, the QC and the QA that pertain to equipment used within the laboratory and how to control for potential contaminants, which reside within the laboratory. This review provides evidence to indicate the need for laboratory personnel to monitor quality improvement issues on a continuous basis. Personnel must be willing to change as improvements in technology occur in order to meet the ever-evolving demands of a more difficult patient population. Suggestions for meeting these demands are offered.
