The effect of Galleria mellonella hemolymph polypeptides on Legionella gormanii.

Abstract: Among Legionella species, which are recognized to be pathogenic for humans, L. gormanii is the second prevalent causative agent of community-acquired pneumonia after L. pneumophila. Anti-L. gormanii activity of Galleria mellonella hemolymph extract and apolipophorin III (apoLp-III) was examined. The extract and apoLp-III at the concentration 0.025 mg/ml caused 75% and 10% decrease of the bacteria survival rate, respectively. The apoLp-III-induced changes of the bacteria cell surface were analyzed for the first… Show more

“…The caterpillar larva (also called wax worms) of the Lepidopteran greater wax moth ( Galleria mellonella ) has become a widely adopted insect model to study several human pathogens including Listeria spp, 26 Streptococcus , 27 Legionella , 28 Campylobacter , 29 Yersinia , 30,31 Pseudomonas , 32-34 Enterococcus , 35 Staphylococcus , 36 Candida , 37,38 Aspergillus 39-41 and other bacterial and fungal pathogens. 42-45 The life cycle of this organism comprises a larval stage that transforms within 6–8 weeks into pupae and finally into moth.…”

“…In addition, ApoLp-III is involved in lipid transport and immunity. Recently, the protein was recovered by methanol extraction, purified and evaluated against three Legionella species: L. dumoffii, L. pneumophila , and L. gormanii ( Palusinska-Szysz et al, 2012 ; Chmiel et al, 2014 ; Zdybicka-Barabas et al, 2014 ). Antimicrobial assays demonstrated a difference in susceptibility among Legionella species.…”

“…Our previous study showed that L. dumoffii cultured in the presence of choline was more sensitive to the antimicrobial peptide defensin and apolipophorin III (apoLp-III) isolated from Galleria mellonella hemolymph [ 11 ]. ApoLp-III, i.e., an insect homolog of human apolipoprotein E exerted a bactericidal effect on L. gormanii cells at an 8-fold lower concentration, compared to L. dumoffii , possibly reflecting differences in the cell surface properties [ 12 ].…”
