Studies were conducted to explore hormonal and nutritional factors that might be involved in the regulation of retinol-binding protein (RBP) synthesis and secretion by the liver. The studies employed primary cultures of hepatocytes from normal rats. When cells were cultured in Dulbecco's modified Eagle's medium alone, a high rate of RBP secretion was observed initially, which declined and became quite low by 24 hr. Supplementing the medium with amino acids maintained RBP and albumin secretion at moderate (but less than initial) rates for at least 3 days. Further addition of dexamethasone maintained the production and secretion rates of RBP, transthyretin, and albumin close to the initial rates for up to 3-5 days in culture. The effects of dexamethasone were not rapid and were not specific for RBP; half-maximal effects were seen at 10(-9) to 10(-8) M levels. Hormonally treated hepatocytes produced and secreted RBP, transthyretin, and albumin at both absolute and relative rates similar to physiological values, as estimated from rates reported by others from studies in vivo (with both rats and humans) and with perfused livers. Glucagon addition partially maintained the secretion rates of these 3 proteins, but less effectively than did dexamethasone. A number of other hormones, added singly or in combination, did not affect RBP production or secretion. Addition of retinol to the cultured normal hepatocytes was without effect upon RBP secretion. These studies show that supplementing the culture medium of hepatocytes with amino acids and dexamethasone maintains RBP production and secretion for several days. In normal hepatocytes, with ample supply of retinol available within the cell, addition of exogenous retinol does not appear to influence RBP metabolism or secretion by the cells.

Effects of dexamethasone on albumin and collagen gene expression in primary cultures of adult rat hepatocytes

To further our studies on collagen gene expression, we have evaluated the molecular basis for the finding that steroids decrease collagen synthesis in cultured hepatocytes. We studied the effects of dexamethasone on primary cultures of adult rat hepatocytes grown on tissue culture plastic in either serum-supplemented medium or a serum-free hormonally defined medium. Cells were plated and allowed to attach for 24 hr in a mixture of serum-supplemented medium + hormonally defined medium. Cultures were then fed every 24 hr for 4 days under 1 of 4 conditions: serum-supplemented medium, serum-supplemented medium + dexamethasone, hormonally defined medium or hormonally defined medium + dexamethasone. On the fifth day, RNA was extracted. Dexamethasone did not affect the amount of RNA isolated; nor did it influence the quantitative translation of the mRNA in the rabbit reticulocyte lysate mRNA-dependent system. Employing hybridization analysis, dexamethasone resulted in increased albumin mRNA content in hepatocytes grown in serum-supplemented medium but had no affect on hormonally defined medium, and decreased type I in collagen mRNA content in cells grown in either serum-supplemented medium or hormonally defined medium. In cells cultured in hormonally defined medium, the beta-actin and procollagen mRNA levels were lower than those in serum-supplemented medium, but albumin mRNA levels were higher, and in fact equivalent to those in vivo. beta-Actin mRNA levels were not affected by dexamethasone in either serum-supplemented medium or hormonally defined medium.(ABSTRACT TRUNCATED AT 250 WORDS)

The effect of hydrocortisone on fibronectin synthesis was investigated in cultured skin fibroblasts. Confluent cells were treated with hydrocortisone (10(-7) M to 10(-5) M) for 2 days and labeled with [3H]proline for 24 h. Fibronectin levels in both the culture medium and the cell layer were studied by gelatin-Sepharose affinity chromatography and SDS-polyacrylamide gel electrophoresis. In control cultures of human fetal skin fibroblasts, fibronectin constituted 8% of the total labeled proteins in the medium. The proportion of fibronectin increased to 13.1% at 10(-7) M hydrocortisone, 15.5% at 10(-6) M and to 19.4% at 10(-5) M. The proportion of fibronectin associated with the cell layer remained at 2-3% of total [3H]proline-labeled proteins and did not increase with hydrocortisone exposure. The stimulating effect of hydrocortisone on medium fibronectin was also demonstrated in cultured human newborn foreskin fibroblasts and in rabbit skin fibroblasts.
