The 50KP was resuspended in the homogenization buffer without mercaptoethanol (total volume of 0,133 ml g~^ original fresh weight of plant material). The material obtained in this way was similar to a combined 21KP and 50KP fraction as described by others (Britz et al, 1979, Widell 1980, Goldsmith et al, 1980, Leong and Briggs 1981, Caubergs and De Greef 1981, Aliquots of 2 ml of this membrane suspension were layered onto a 30 ml linear sucrose density gradient of 25 to 45% (w/w) sucrose dissolved in the MES-buffer, The sucrose gradient was centrifuged at 100 000 g in a 6x38 ml swing-out rotor (MSE) for 16 h. The gradients were fractionated in 1,5 ml portions. Those membrane fractions, containing LIAC activities (fractions 5 to 20) were pooled, diluted with 3 times the volume of sucrose-free MES buffer, pelleted and resuspended in 5 ml of the same buffer.…”
