The effect of cilengitide in combination with irradiation and chemotherapy in head and neck squamous cell carcinoma cell lines

Abstract: Cilengitide in combination with cisplatin and irradiation may be a feasible option for the treatment of patients with head and neck cancer. However, further investigations are required to understand the exact mechanism that leads to synergistic cytotoxicity.

“…The most prominent compound targeting integrins might be cilengitide, a cyclic RGD peptide that targets integrins αvβ3 and αvβ5 [214]. We and other groups could show in vitro and in vivo that this peptide inhibits attachment and invasion of different tumor cells [215,216,217,218,219,220,221]. However, cilengitide failed to provide benefit in a clinical trial phase III (CENTRIC study) [222].…”

“…Clonogenic assays were performed as previously described (12). Briefly, 3-15x10 2 cells were seeded into 6-well plates and allowed to settle for 24 h. Afterwards, the cells were exposed to 5-20 µM TQ and/or irradiated with 2, 4, 6 or 8 Gy.…”

“…Cilengitide in combination with irradiation gave significant inhibition of colony formation [74]. In ex vivo experiments, cilengitide alone had insignificant effect on colony formation from biopsies of HNSCC [75], although the same group later found it did significantly reduce colony formation when cells were seeded on laminin [76].…”
