The Effect of Cigarette Smoke Exposure and Ascorbic Acid Intake on Gene Expression of Antioxidant Enzymes and Other Related Enzymes in the Livers and Lungs of Shionogi Rats with Osteogenic Disorders

Abstract: Cigarette smoking causes many chronic diseases but is a preventable risk factor in developing countries. However, it may be possible to relieve the smoke-induced damage by increasing the protective defense system. As vitamin C intake reduces smoking risk, it is recommended that smokers should take more vitamin C. However, the molecular mechanism of vitamin C intake on smokers has not been thoroughly investigated. We have found there to be suppression of smoke-induced cytochrome P-450 1A1 (CYP1A1) mRNA expressi… Show more

“…The RNA was purified by RNeasy Mini Kit (Quiagen, Valencia, CA) according to the manufacturer's instructions. The reverse transcriptase (RT) mixture (30 Al) containing 2 ml of total RNA was made and incubated at 37jC for 60 min by a previously reported method (Ueta et al, 2003). One ml of the mixture was used for real-time PCR, which was carried out using a LightCycler thermal cycler system with a Light Cycler SYBR Green I kit according to the manufacturer's instructions (Roche Diagnostics, Tokyo, Japan) (Wittwer et al, 1997).…”

“…Those that are available model primarily exposure to pollutants soon after infection, during the incubation phase, and demonstrate potentiation of the disease process (Becker and Soukup 1999; Harrod et al 2003; Lambert et al 2003). Epidemiologic studies of childhood asthma exacerbation by air pollution exposure examined a variety of acute windows of exposure and found several exposure periods/lags important for adverse events ranging from 0 to 5 days (Gouveia and Fletcher 2000; Lee et al 2002; Romieu et al 1995; Sunyer et al 1997).…”
