Diabetes mellitus is a chronic syndrome affecting many systems of the body and may be considered as a leading cause of death globally. To compare the efficacy of Stevia rebaudiana and Aspartame on hepato-renal functions, sixty adult male albino rats were divided into 6 groups; control, Stevia, Aspartame, diabetic, diabetic-treated with Stevia and diabetic-treated with Aspartame. Food consumption, body weight, blood glucose, serum aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), urea and creatinine levels were determined. Hepatic malondialdehyde (MDA), glutathione (GSH) and bcl-2 levels were assessed as well as histological examination for liver and kidney sections were performed. Treatment of diabetic rats with Stevia leaf extract or Aspartame significantly lowered food consumption and serum blood glucose. Serum blood glucose was significantly decreased by 38% with Stevia treatment and 50% with Aspartame. Stevia treatment of diabetic rats significantly lowered the serum level of ALT by 62%, AST by 57%, ALP by 41%, urea by 16% and creatinine by 65%. Hepatic MDA was reduced by 50% in diabetic group treated with Stevia and hepatic GSH and Bcl-2 levels were significantly elevated. In contrast, Aspartame did not change these parameters. Histopatholgically, diabetic rats treated with Stevia showed improvement of liver and renal tissues, while Aspartame treatment augmented alloxan induced tissue destruction. Aspartame was more effective hypoglycemic agent than Stevia in diabetic rats but Stevia showed protective effects on the liver and kidney functions. Stevia rebaudiana may be therapeutically beneficial as hepato-renal protective agent for those suffering from diabetes mellitus.

Objectives: This study aimed to elucidate the possible protective effect of simvastatin (SIM) pretreatment on stress-induced gastric ulcer in rats. Materials and Methods:Gastric ulcer was produced in Sprague-Dawley rats (250-300 g) by cold-restraint stress. SIM (10 mg/kg/day; per oral) or saline was administered for 21 days prior to stress. On day-21, a group of animals was treated with the non-selective nitric oxide synthase (NOS) inhibitor L-NAME (50 mg/kg; intraperitoneally) or the non-selective cyclooxygenase (COX) inhibitor indomethacin (Indo; 5 mg/kg; subcutaneously) prior to SIM. The stomachs were examined macroscopically and microscopically and stored for biochemical analyses. Results:The severity of the lesions of the stress group was decreased by SIM, but this was not altered significanty by L-NAME or Indo. Stress increased gastric myeloperoxidase activity comparised to control level (p<0.01); however, SIM did not cause a significant change on this parameter. Stress increased gastric chemiluminescence levels (p<0.001) which were reversed by SIM (p<0.001) and this effect continued in L-NAME-or Indotreated animals.Conclusion: SIM pretreatment of rats with cold-restraint stress provided protection against gastric lesion formation via supression of oxidants derived from sources other than neutrophils without the involvement of NOS and COX systems.Keywords: Ulcer, Nitric oxide synthase, Cyclooxygenase, Rat, Stress ÖZ Amaç: Bu çalışmada sıçanlarda stres ile uyarılan mide ülserinde simvastatin'in (SİM) olası koruyucu etkisini değerlendirmek amaçlanmıştır. Gereç ve Yöntem:Sprague-Dawley sıçanlarda (250-300 gr) soğuk-kısıtlama stres uygulaması ile mide ülseri oluşturuldu. Stres öncesi 21 gün süreyle SİM (10 mg/kg/day; oral yolla) veya fizyolojik tuzlu su (1 ml; oral yolla) uygulandı. Bir grup sıçan 21. günde SİM öncesi selektif olmayan nitrik oksit sentaz (NOS) inhibitörü L-NAME (50 mg/kg; intraperitoneal) veya selektif olmayan siklooksijenaz (COX) inhibitörü indometazin (İndo; 5 mg/kg; subkutan) verildi. Mideler makroskopik ve mikroskopik olarak incelendi ve biyokimyasal analizler için saklandı. Bulgular:Stres grubunda lezyonların şiddeti SİM ile azaldı ancak L-NAME veya İndo ile anlamlı bir değişiklik göstermedi. Stres mide miyeloperoksidaz aktivitesini kontrol düzeyine kıyasla artırdı (p<0.01); ancak, SİM bu parametre üzerinde anlamlı bir değişikliğe neden olmadı. Stres mide kemiluminisans düzeylerini artırdı (p<0.01); bu etki SİM ile geri döndürüldü (p<0.001); L-NAME veya İndo ile tedavi edilen hayvanlarda ise devam etti.Sonuç: Soğuk-kısıtlama stresine maruz bırakılan sıçanlara SIM öntedavisi mide lezyon oluşumunda NOS ve COX sistemlerinden ayrı mekanizmalarla nötrofil dışı hücrelerden kaynaklanan oksidanları baskılayarak koruma sağlamaktadır.

The aims of the present study were to develop a colon-specific gel formulation of melatonin with sodium alginate and to evaluate its in vitro characteristics and intracolonic performance on oxidative stress parameters, such as nitric oxide (NOx), malondialdehyde (MDA) and glutathione (GSH) levels in rats with acetic acid-induced colitis. The melatonin-alginate gel formulations were prepared and their physico-pharmaceutical properties were determined. Formulation M5, which contained 3% of sodium alginate and 20% polyethylene glycol, was used for in vivo studies. The in vivo studies were conducted in rats with acetic acid-induced colitis. NOx, MDA and GSH levels were determined and histological investigations were performed. It was found that formulation M5 was the most suitable formulation for the colon-specific melatonin gel, in terms of pH, viscosity, drug release and mucoadhesion properties. The MDA levels in the tissues of Group 2 (treated with an intracolonic gel formulation without melatonin) were found to be significantly higher than in Group 1 (the untreated group). NOx levels decreased with the intracolonic and systemic melatonin treatment in the colitis-induced rats. Neither intracolonic nor intra-peritoneal (IP) melatonin treatment affected GSH levels. The epitelization of the colon tissues in groups administered with intracolonic melatonin, IP melatonin, and the intracolonic gel formulation without melatonin was much better than that found in the untreated group. It was concluded that melatonin participated in various defense mechanisms against the colonic inflammatory process, and that the dose, route and formulation type were the most important parameters in the effectiveness of melatonin.
