The effect of alginate-gelatin encapsulation on the maturation of human myelomonocytic cell line U937

Abstract: Today, many attempts have been collected in the field of tissue engineering for reconstitution of injured bone marrow capacity by transplantation of functional cell source. By having a three-dimensional condition, microcapsules are appropriate candidates for cells transplantation to target sites. Here, we examined the effect of alginate-gelatin microcapsules on functional maturation of human myelomonocytic cell line U937 after 7 days in vitro. U937 cells were encapsulated by the mixture of alginate-gelatin and… Show more

“…Cells were incubated for 30 min at different temperatures (4 and 37 • C) after been mixed within the bioink, consisting of A1.8 G3. For releasing the embedded cells, 0.1 M sodium citrate (pH = 7.4) was used as a chelating agent at 37 • C until the gel was dissolved, as previously described (Nemati et al, 2019). In the control group (C), cell viability was measured without the 30 min incubation step and mixture with bioink.…”

“…The U937 cells grown in suspension are characterized by a round shape, short microvilli, and a large beam-shaped nucleus (Pagliara et al, 2005;Tenuzzo et al, 2008). These monocytic cells bear the potential of differentiating into macrophages or dendritic cells; in response to a number of stimuli adopt the morphology and physiological characteristic of macrophages and thus represent a precursor of mononuclear phagocyte system (Kigerl et al, 2009;Italiani and Boraschi, 2014;Nemati et al, 2019).…”

“…To overcome these limitations, protective microenvironments with proper biochemical and mechanical cues are essential to dictate specific cellular behavior and function [ 28 , 29 ]. In recent years, tissue engineering, a branch of interdisciplinary science, can help researchers and clinicians with accelerated tissue healing using de novo technological approaches consisting of specific cell populations, growth factors, and scaffolds [ 30 , 31 ]. In this article, recent data associated with the application of varied scaffold types for the regeneration of injured muscle tissue was investigated.…”
