The Effect of Age on the Synthesis of Two Heat Shock Proteins in the HSP70 Family

Abstract: Hepatocytes isolated from male F344 rats were exposed to elevated temperatures (40 degrees C to 45 degrees C), and the incorporation of [35S]-L-methionine into proteins was measured from fluorograms of two-dimensional polyacrylamide gels. The synthesis of two proteins was induced by temperatures of 40 degrees C to 42.5 degrees C; however, 45 degrees C inhibited the synthesis of all proteins. Based on their apparent molecular weights and pI values and their recognition by a monoclonal antibody to the HSP70 gene… Show more

“…Hsf-1 is the transcription factor for Hsp70 and therefore regulates its synthesis in response to stress. Consistent with this, it has been shown in both rat hepatocytes (Wu et al, 1993) and in human T-lymphocyte cultures (Effros et al, 1994) that with age there is a decrease in Hsp70 synthesis. However, a recent study by Hsu and colleagues contradicts this hypothesis since Caenorhabditis elegans forced to over express the hsf-1 gene exhibited 40% greater life-span than wild type animals (Hsu et al, 2003).…”

“…Aging usually lowers the expression of antioxidant enzymes and stress protein expression. This loss can be modulated through interventions with diet or exercise [58,62,118]. One effect of the combined treatment EA was a significant increase in the expression of inducible heme oxygenase (HO-1) also known as HSP32.…”

“…Aging is associated with altered expression of heat shock protein gene family members such as Hsp70 and Hsc70, 28,29 whereas HFDR significantly induced their mRNA level (Table 2). In addition, Lamp2a protein expression was twofold higher in restricted than AL-fed mice ( p = 0.022; Fig.…”
