The ectopic expression of the wheat Puroindoline genes increase germ size and seed oil content in transgenic corn

Abstract: Plant oil content and composition improvement is a major goal of plant breeding and biotechnology. The Puroindoline a and b (PINA and PINB) proteins together control whether wheat seeds are soft or hard textured and share a similar structure to that of plant non-specific lipid-transfer proteins. Here we transformed corn (Zea mays L.) with the wheat (Triticum aestivum L.) puroindoline genes (Pina and Pinb) to assess their effects upon seed oil content and quality. Pina and Pinb coding sequences were introduced … Show more

“…The Puroindoline D1 genes have a dramatic impact on kernel texture, a significant effect on processing quality and may play a role in plant disease defense (Chen et al 2007;Feiz et al 2009;Morris 1997, 1998;Lillemo and Morris 2000;Ma et al 2009;Zhang et al 2010Zhang et al , 2011 Morris et al 2001;Xia et al 2005). In this study, we analyzed the frequency of PinaD1b and Pinb-D1b among four hard wheat groups (hard red winter, hard white winter, hard red spring and hard white spring), and found that 94 of 101 hard winter wheats (red and white) carried Pina-D1b (Table 3).…”

“…An additional effect was reported by Zhang et al (2010) who found that constitutive expression of the puroindolines in corn increased embryo size and total seed oil content. The mechanism of embryo size increases in corn or of polar lipid content in wheat is not clear but both may result directly from the affinity of puroindolines for lipids (Dubreil et al, 1997;Feiz et al, 2009b).…”

“…An example of this novel approach to engineering carbon flow includes the expression of a lipid transfer protein relative, puroindoline. When expressed in maize, puroindoline caused a 25% increase in total seed oil content …”
