Dastarcus helophoroides Fairmaire (Coleoptera: Bothrideridae) is an effective predatory beetle of larvae and pupae of several cerambycid beetles including Monochamus alternatus and Anoplophora glabripennis. Electroantennography (EAG) and a dynamic two-choice olfactometer were respectively used to measure the antennal and behavioral responses of both sexes to selected volatile compounds. Female and male D. helophoroides exhibited similar EAG and behavioral responses. Significant dose-dependent EAG responses in both sexes were elicited by nonanal, octanal, cis-3-hexenol, 3-carene, (R)-(+)-α-pinene, (S)-(-)-α-pinene, (R)-(+)-limonene and (S)-(-)-limonene. Female and male beetles were repelled at high concentration by cis-3-hexenol and (S)-(-)-limonene, respectively. Both sexes of D. helophoroides were significantly attracted to nonanal, cis-3-hexenol, 3-carene and (R)-(+)-limonene even at low concentrations. These compounds might be used either individually or in mixtures for developing biological control methods to attract this predatory beetle into forest stands threatened by cerambycid beetles.

BACKGROUND: Pine wilt disease (PWD) is responsible for extensive economic and ecological damage to Pinus spp. forests and plantations worldwide. PWD is caused by the pine wood nematode (PWN, Bursaphelenchus xylophilus) and transmitted into pine trees by a vector insect, the Japanese pine sawyer (JPS, Monochamus alternatus). Host infection by PWN will attract JPS to spawn, which leads to the co-existence of PWN and JPS within the host tree, an essential precondition for PWD outbreaks. Through the action of their metabolites, microbes can manipulate the co-existence of PWN and JPS, but our understanding on how key microorganisms engage in this process remains limited, which severely hinders the exploration and utilization of promising microbial resources in the prevention and control of PWD. RESULTS: In this study we investigated how the PWN-associated fungus Aspergillus promotes the co-existence of PWN and JPS in the host trees (Pinus massoniana) via its secondary metabolite, sterigmatocystin (ST), by taking a multi-omics approach (phenomics, transcriptomics, microbiome, and metabolomics). We found that Aspergillus was able to promote PWN invasion and pathogenicity by increasing ST biosynthesis in the host plant, mainly by suppressing the accumulation of ROS (reactive oxygen species) in plant tissues that could counter PWN. Further, ST accumulation triggered the biosynthesis of VOC (volatile organic compounds) that attracts JPS and drives the coexistence of PWN and JPS in the host plant, thereby encouraging the local transmission of PWD. Meanwhile, we show that application of an Aspergillus inhibitor (chiricanine A treatment) results in the absence of Aspergillus and decreases the in vivo ST amount, thereby sharply restricting the PWN development in host. This further proved that Aspergillus is vital and sufficient for promoting PWD transmission. CONCLUSIONS: Altogether, these results document, for the first time, how the function of Aspergillus and its metabolite ST is involved in the entire PWD transmission chain, in addition to providing a novel and long-term effective nematicide for better PWD control in the field.

For animals that live in association with humans, a key ecological question is how anthropogenic factors influence their life history. While major negative effects are obvious, subtle non-lethal responses to anthropogenic stimuli may provide insight into the features that lead to the success of species that thrive in habitats heavily impacted by humans. Here we explored the influence of the herbicide atrazine on various life history traits of a wolf spider that thrives in agroecosystems where it is commonly applied. We found that exposure delayed maturation and increased the probability of having molting errors. Atrazine also decreased the probability of producing an egg sac after mating, but increased the average mass of the initial egg sacs that were produced while not impacting the average number of eggs inside. The total number of eggs produced from a single mating on the other hand, was increased in the presence of atrazine through the production of multiple egg sacs. Finally, adult lifespan was shortened with exposure to atrazine. These results suggest that the atrazine based herbicides that are routinely applied to agricultural fields result in altered life history traits, potentially through endocrine disruption, that may counteract one another to allow persistence.
