The E3 ubiquitin ligase LNX1p80 promotes the removal of claudins from tight junctions in MDCK cells

Abstract: The structural continuity of tight junctions (TJs) is consistently maintained even when epithelial cells divide and move within the cellular sheet. This process is associated with dynamic remodeling of TJs by coordinated internalization and generation of claudin-based TJ strands, but the molecular mechanism behind the regulated turnover of TJs remains largely unknown. In this study, we identified the p80 isoform of the E3 ubiquitin ligase ligand of Numb-protein X1 (LNX1p80) as a protein binding to claudin-1. I… Show more

“…The constitutive turnover of claudin-2 is thought be mediated by an E3 ubiquitin ligase that targets claudins for subsequent lysosomal degradation (16). However, the signaling pathways that regulate claudin-2 turnover have not been established.…”

“…1C) (Dho et al, 1998;Rice et al, 2001). However, in contrast to LNX2, the function of LNX1p80 as E3 ligase has been partially explored (Kansaku et al, 2006;Takahashi et al, 2009). For these reasons, we decided to test whether LNX1 is also a CD8a binding protein.…”

“…Next, we examined whether LNX1p80 was able to ubiquitylate CD8a, as previously described for claudin (Takahashi et al, 2009), and whether also LNX2 was endowed of ubiquitylation activity. For this purpose, we coexpressed CD8a, HA-tagged ubiquitin, and GFP, GFP-LNX1p80 or GFP-LNX2 in HEK293 cells (Fig.…”
