The Dystonia Coalition: A Multicenter Network for Clinical and Translational Studies

Abstract: Dystonia is a movement disorder characterized by sustained or intermittent muscle contractions causing abnormal postures, repetitive movements, or both. Research in dystonia has been challenged by several factors. First, dystonia is uncommon. Dystonia is not a single disorder but a family of heterogenous disorders with varied clinical manifestations and different causes. The different subtypes may be seen by providers in different clinical specialties including neurology, ophthalmology, otolaryngology, and oth… Show more

“…Data for these analyses were obtained from the Dystonia Coalition, an ongoing multicenter international project aimed at delineating the clinical features and evolution of isolated dystonias. Methodological details for this study, 9 and clinical details for portions of this cohort have been presented elsewhere. [10][11][12] Briefly, subjects were recruited by 57 different specialists in movement disorders at 40 sites across North America, Europe, and Australia.…”

“…Data from the DC database were collected and analyzed for 727 OMD subjects enrolled across 26 international sites from 2011 to 2019 ( http://clinicaltrials.gov/show/NCT01373424 ) ( 17 ). To be included in the DC database subjects had to be diagnosed with isolated dystonia of any type (focal, segmental, multifocal, generalized).…”

“…The data from the DC also provides new information for a large number of OMD cases (n = 727). Since multiple centers used the same protocols for evaluation, these data reflect the most consistent information from the largest number of different centers (17). These data also include quantitative rating scale data for all body regions.…”

“…We retrospectively analyzed clinical data and video recordings from 206 CD patients enrolled across 10 North American academic centers in a cross‐sectional rating scale validation study ( https://clinicaltrials.gov/ct2/show/NCT01373424 ) 24 previously conducted by the Dystonia Coalition. 25 All participants were evaluated in person by movement disorders neurologists with expertise in dystonia, and only those with isolated (primary) dystonia were enrolled into the study. Participants were treated with periodic BoNT injections and a variety of medications, including benzodiazepines, GABAergics, beta adrenergic blockers, dopaminergics, and anti‐cholinergics.…”
